The note specifically evaluates ZyGEM’s prepGEM(TM) Tissue DNA extraction kits used in PCR assays for genotyping mice, a commonly-performed biomedical research application.
In the application note, ZyGEM researchers describe their experience extracting DNA and determining precise genotypes from mouse tail samples. Researchers performed extractions comparing ZyGEM’s prepGEM(TM) Tissue kit with two other commercially available kits with similar protocols. They then amplified a relevant segment of DNA using PCR reagents supplied in the kits and also with an external amplification system. The prepGEM(TM) Tissue extractions and amplifications yielded superior results based on both qualitative and quantitative PCR comparisons of the amplified DNA.

The results included in this application note show that in a PCR assay gel analysis, the prepGEM(TM) Tissue kit extractions yielded readily amplified DNA with no migration distortion or smearing, while the other two commercial kits yielded DNA with migration distortion or low band intensity, indicating a low yield of extracted DNA. The quantitative PCR analysis showed that samples prepared with prepGEM(TM) Tissue yielded over 34 times the amount of DNA produced by the higher-yielding of the two comparison kits.
The prepGEM(TM) Tissue kit is compatible with all PCR detection and amplification reagents, and the stability of the prepGEM(TM) reagents allows convenient storage at 4 degrees C, reducing preparation time and increasing ease of use.
