FragIT Kit contains one spin column for IgG fragmentation (FragIT spin column) and one Fc capturing spin column. The kit is available in three standard sizes; 0.5, 10 and 100 mg. Genovis provides other format on request.
FragIT spin columns come prefilled with FabRICATOR® enzyme immobilized on agarose that allow for easy and quick fragmentation of IgG into F(ab’)2 and Fc fragment. Fragmentation of IgG is typically associated with low yield, heterogeneity of the produced fragments and a cumbersome and time- consuming process. All these shortcomings can now be solved using FragIT columns. After elution from the FragIT column, F(ab’)2 and Fc fragments are separated using a Fc capturing spin column. This column contains anti Fc IgG molecules conjugated to agarose. The F(ab)2 fragments are easily eluted at a very high yield.

FabRICATOR®
Genovis has developed FabRICATOR, to rapidly generate precise F(ab’)2 fragments. FabRICATOR is a partially modified and his-tagged IdeS enzyme* that cleaves IgG in a singular defined site below the hinge region leaving an intact F(ab?)2 and two residual Fc fragments. The yield of the reaction is high and the enzyme does not further digest the resulting products. *IdeS is a highly substrate specific cysteine protease originating from Streptococcus pyogenes.
