Pharmaceutical development is an extremely complex process which involves discovery of innovative active ingredients all the way to the launch of a finished product. It is costly and often takes more than a decade before the final success. During this process, hit identification for the discovery of lead compounds are the initial research and largely determines whether the drug development would come out as a success or not.

However, there are also strategies that do not require any prior information on target or ligand, which generally use serendipity-based search strategies in either a given physical or virtual compound subset. With the help of high-throughput screening (HTS), such random hit-identification strategies are made possible.
Mr. Barron Jones further explains these three methods, “NMR spectroscopy is a useful tool in the hit identification, which has been widely applied to detect ligand binding and to monitor enzyme kinetics and inhibition. X-ray crystallography is a technique developed to analyze the atomic and molecular structures of crystals, and the structural information obtained is then used to evolve the fragments to hits, either by growing the fragment or combining structural features from a lot of compounds. HTS, widely used in drug discovery, is a scientific method that can help identify hits, antibodies, or genes that regulate a specific biological pathway.”
