Biotech Support Group Release: Cyclic Nucleotide Phosphodiesterases Enrichment Product Cited in Research on Alzheimer’s Disease

PRLog (Press Release) - Dec 08, 2012 - MONMOUTH JUNCTION, NJ –Biotech Support Group reports on a recent journal article which cites the use of its research consumable product - PDEnRich™. The article explores the mechanism of caffeine and melatonin on preventing cognitive dysfunction in transgenic mouse models of Alzheimer’s disease. In part, it reports the effect of inhibiting cAMP-dependent phosphodiesterase (4) or cGMP-dependent phosphodiesterase (5) on melatonin protection of mitochrondrial function, citing the use of the PDEnRich™ kit for cyclic nucleotide phosphodiesterase (PDE) enrichment. Moreover the effect of caffeine and cyclic nucleotide levels on melatonin signaling was observed in mitochondria by independent mechanisms that do not involve adenosine receptor antagonism. The results of this study indicate that melatonin restores mitochondrial function much more potently than caffeine in transgenic mouse and cell models of Alzheimer’s disease.

“This article demonstrates the importance of understanding the mechanism of action of phosphodiesterase inhibitors and so our new methods for enriching phosphodiesterases from natural sources will improve the quality of research in this area. However, because of the instability of cyclic nucleotides, classical substrate affinity enrichment is not an option for these enzymes, so we worked around that problem to develop PDEnrich™. As a result, this product can be a crucial first step to prospect into the structure and function of phosphodiesterase variants.” states Swapan Roy, Ph.D., President of Biotech Support Group.

PDEnRich™ is derived from a silica-based NuGel™ multi-dimensional surface library and optimized for the enrichment of phosphodiesterase activity. The standard prep protocol starts with 100 µl of tissue/cell extracts, or approximately 0.5 mg total protein, but the process can be scaled up or down to accommodate different sample volumes and protein concentrations. The kit includes all necessary reagents for immediate use.
