Creative Biolabs Released Two Hybrid Services

With the common efforts of its scientists, Creative Biolabs released two hybrid services.

As we know, an understanding of how proteins interact within the cell is a key part of gene functional analysis and potential drug target discovery. The two-hybrid system is a very powerful molecular genetic tool which investigates protein to protein interactions in vivo. This system can be used to study the interaction between two proteins which are expected to interact and find proteins interacting with a protein you have already bait.

Creative Biolabs provides the services on a fee-for-service basis. It is specialized in using Gal4 and LexA based Y2H systems. It is able to perform yeast 2 hybrid screening against integral membrane proteins and membrane-associated/proximal proteins utilizing a modified split-ubiquitin membrane yeast two-hybrid (MYTH) system.

Yeast One-Hybrid Screening

One-hybrid screening in yeast is a powerful method to rapidly identify heterologous transcription factors that can interact with a specific regulatory DNA sequence of interest. In this technique, the interaction between two proteins (bait and prey) is detected via in vivo reconstitution of a transcriptional activator that turns on expression of a reporter gene.

Detection is based on the interaction of a transcription factor (prey) with a bait DNA sequence upstream of a reporter gene. To ensure that DNA binding results in reporter-gene activation, cDNA expression libraries are used to produce hybrids between the prey and a strong trans-activating domain. Compared to biochemical techniques, the advantage of cloning transcription factors or other DNA-binding proteins via one-hybrid screenings is that the procedure does not require specific optimization of in vitro conditions. Its combination of the 1-hybrid and 2-hybrid approaches increases the stringency and reliability of results obtained from 2-hybrid assays alone.
