The proteasome has been validated as an important clinical target in cancer, and Onyx is developing next-generation proteasome inhibitors with a high degree of selectivity, with the goal of increasing therapeutic efficacy and reducing off-target toxicities. Based on results from the Phase 2b 003-A1 study evaluating our lead product candidate in this program in patients with relapsed and refractory multiple myeloma, Onyx has initiated a rolling submission of a New Drug Application (NDA) for potential accelerated approval in the U.S. and initiated two Phase 3 trials to support full registration in the U.S. and Europe.
The Onyx development program also includes an oral proteasome inhibitor that is currently in Phase 1 testing in solid tumors; an immunoproteasome inhibitor with activity in preclinical models of autoimmune disorders; an alpha-folate receptor targeted inhibitor of thymidylate synthase that is in Phase 1 testing; and an oral small molecule cyclin-dependent kinase 4/6 (CDK4/6) inhibitor, now in Phase 2 clinical development, resulting from a collaboration with Warner-Lambert Company, now Pfizer.
