Project Outline:

The goal of this project is to optimize and validate next-generation sequencing (NGS) based assays for the detection of potential adventitious agents in cell banks, cell drug products, and viral vectors.

Adventitious agents are unwanted or unintended biological agents, such as viruses, that may contaminate cell cultures or final cell therapy products during the manufacturing process. Detecting these agents is crucial for ensuring the safety and quality of cell therapy products. NGS technology, known for its high-throughput and high-resolution sequencing capabilities, allows for comprehensive and sensitive detection of a broad range of adventitious agents within a single assay. Incorporating NGS-based adventitious agent testing into the development and manufacturing processes of cell therapy drug products can significantly enhance safety, quality control, and compliance with regulatory standards, ultimately contributing to the successful translation of these therapies from the laboratory to clinical application.

The intern will be responsible for supporting the assay development, conducting experiments, analyzing data, and assisting in the overall qualification process and work onsite (Cambridge, MA) The project aims to enhance the safety and quality control of cell drug products through robust adventitious agent testing using NGS technology.

How you will contribute:
  • Work closely with other team members to optimize and validate NGS-based assays for adventitious agent testing in cell therapy samples.

  • Participate in assay development, including protocol design, reagent preparation, and optimization of sequencing parameters.

  • Assist in the preparation and processing of cell therapy samples for NGS-based adventitious agent testing.

  • Perform data analysis using bioinformatics tools to identify potential adventitious agents and assess the quality of sequencing data.

  • Maintain detailed and accurate records of experiments, protocols, and results.

  • Summarize findings in regular reports and presentations for internal team meetings.

  • Contribute to the implementation of quality control measures to ensure assay reproducibility and accuracy.

  • Work on troubleshooting and resolving technical issues related to assay performance.

  • Collaborate with cross-functional teams to integrate assay results into broader cell therapy programs.
