The Preformulation group is an integral part of the Biologics Drug Product Development organization at AbbVie. The primary function of the Preformulation team is to screen discovery-stage biologic drug candidates for developability and to help select leads with optimal biophysical and biochemical properties suitable for late-stage development, improving the likelihood of success. Our strong pipeline consists of highly varied biologic drug classes, including mAbs, bispecifics, antibody-drug conjugates, fusion proteins, and AAV gene therapies against diverse therapeutic targets. We have an exciting opportunity for a Principal Research Scientist I in the Biologics Drug Product Development (Preformulation) team, based in Worcester, Massachusetts.

 

Key responsibilities include:

·       Responsible for overseeing the successful advancement of discovery pipeline candidates as well as driving biologics drug product research initiatives.

·       Designs experiments and strategies to efficiently evaluate physicochemical and biophysical properties of biologics and identify liabilities that impact product development of new biologic entities (e.g., antibodies, antibody-drug conjugates, bispecific antibodies, fusion proteins). Selects biologic candidates with desired characteristics for further product development. 

·       Serves as project lead for the Preformulation group, interacting with team members from discovery and development groups to optimize a matrixed workflow of discovery molecules toward new pipeline development. Learns about diverse disease areas and new biological therapeutic initiatives and fosters effective collaborations with key partner teams.

·       Uses advanced knowledge of formulations, biophysics, biochemistry, and CMC to streamline development of new biologics.

·       Takes initiatives to identify new methods and seeks collaborations to pursue research ideas and scientific questions that may arise during Preformulation studies. Drafts technical documents and patent applications. Writes manuscripts for publication in peer-reviewed journals. Attends external conferences for professional development. Communicates ideas and plans to colleagues at all levels of the organization. 

·       Mentors junior colleagues and builds a highly effective and collaborative team. Helps maintain a transparent work environment. Emphasizes efficient work habits and detailed attention to data documentation. Manages product timelines and resources.

·       Executes a strong laboratory-based workplan to achieve results. Seeks cutting-edge methods and instruments to advance goals.


Basic qualifications include:

·       Ph.D. (6+ years), or MS (12+ years) with multi-disciplinary scientific experience in Biophysics, Biochemistry, Pharmacy, Biopharmaceutics, or related discipline. Strong laboratory and technical expertise in biologics and formulation.

·       Advanced knowledge and experience in biophysics, physicochemical protein methods, discovery research and biologic process and product development. 

·       Detailed familiarity with protein and small-molecule analytical methods including liquid chromatography (SEC, ion-exchange, hydrophobic-interaction), mass spectrometry, analytical ultracentrifugation, UV-VIS absorbance and fluorescence spectroscopy, differential scanning calorimetry, particle size and charge measurement, dynamic light scattering, capillary electrophoresis, gel electrophoresis, Western blotting. Experience with high-throughput methods and automation. 

·       Experience with diverse biologic therapeutics, e.g., mAbs, bispecifics, fusion proteins, antibody-drug conjugates.

·       In-depth knowledge of protein formulation and solution biophysics including colloidal interactions, viscosity and thermal (conformational) stability measurements. Willing to learn new technologies and knowledge areas as needed. 

·       Computational chemistry knowledge and protein structural modeling experience.

·       Detailed knowledge of biologics and track record of product development. Experience and interest in interpretation of ligand-binding assays, bioactivity measurements, and pharmacokinetics data. 

·       Excellent communication skills. Demonstrated proficiency in writing technical documents and peer-reviewed publications.

·       Leadership experience of leading and mentoring junior colleagues.

·       Hands-on laboratory operation and organizational skills are highly desired.
