Job Description

About the role:

At Takeda, we are committed to lifelong learning.

To that end, Takeda's summer internship program blends real world experience with an extensive overview of the pharmaceutical industry. Knowledgeable mentors will provide guidance as you gain professional hands-on experience to start your career or further develop in your expertise.

The summer internship program is a full-time commitment of 12 weeks in length and offers a unique perspective into a world-class pharmaceutical company. Our internship program also provides you the opportunity to network with people at Takeda through various planned events and activities.

Project Outline:

Synthetic small interfering RNA (siRNA) therapeutics provide a unique modality designed to selectively target and degrade specific messenger RNA (mRNA) that are involved in relevant disease pathways. One of the benefits of this modality is that it allows for selective targeting of any mRNA based on the target sequence. Because siRNA therapeutics cannot reach the site of action directly by diffusion, they must rely on different delivery vehicles that require encapsulation in a lipid nanoparticle or conjugation to a ligand that can internalize in a tissue of interest. Takeda is currently pursuing several siRNA therapies and is in need of a reliable human efficacious dosing regimen prediction for this modality to support progression of these therapies to the clinic. While there are a few FDA-approved siRNA therapeutics, there is still a poor understanding of the translation of efficacious dosing regimen from discovery to the clinic. In part, this may be due to the fact that siRNA exposure has been shown to be dependent on route of administration and the rate of siRNA internalization, which may be species dependent.

To address this gap, Takeda’s DMPK group is searching for a highly motivated candidate, who will compile, integrate, and analyze proprietary and published in vitro, in vivo, and clinical data and leverage various modeling approaches to help inform our understanding of the pharmacokinetics (PK) and tissue distribution, uptake kinetics, and intracellular trafficking of siRNAs across species. The candidate will build a mathematical model that captures the effects of the route of administration and internalization on the overall exposure of siRNAs. This work will help to enable human PK projections from preclinical studies for first-in-human dose predictions. The candidate will be part of a working group comprised of members from DMPK and Quantitative Solutions to address the human dose prediction questions across various siRNA programs.
