ShapeTX is looking for a Senior Research Associate / Associate Scientist / Senior Associate Scientist with a background in molecular biology and mammalian cell culture to join our Technology Development team in Boston, MA. In this position, the successful candidate will advance technology development across ShapeTX's novel gene therapy platforms, in particular (but not limited to) RNA editing. This role would use a design-build-test synthetic biology approach along with high-throughput, multiplexed assays to improve gene therapy constructs.  At a minimum, the candidate must have experience in plasmid cloning and culturing cell lines. The ideal candidate will have experience in next-generation sequencing, cell line engineering, and single-cell analysis. Attention to detail and interest in learning new cutting-edge techniques is essential.

ShapeTX has developed a suite of technology platforms that broadly enable RNA targeting, RNA editing and RNA replacement for diseases with high unmet need. By leveraging enzymes already present in human cells such as ADAR (adenosine deaminase acting on RNA), RNA editing eliminates the immunogenicity and malignancy risks associated with DNA-based editing platforms such as CRISPR. RNA editing requires no foreign proteins, just the delivery of a single guide RNA that can be gene-encoded and packaged in AAV for precise delivery. RNA editing can be fine-tuned to achieve the desired biological outcome and, together with our AAVidTM capsid discovery platform and SquareBio stable cell line manufacturing platform, we can target diverse tissues and overcome current industry manufacturing challenges. Roles and Responsibilities

  • Execute high throughput screening methods in cultured cell lines to evaluate guide RNAs for therapeutic RNA editing, including: gene synthesis; cloning plasmid libraries; mammalian cell culture and transfection; RNA purification; amplification of target genes by qPCR and droplet digital PCR; Next-Generation Sequencing; single-cell barcoding
  • Design and plan well-controlled experiments. Seek opportunities to improve our current protocols and workflows. Troubleshoot as needed to find creative solutions.
  • Assist with lab operations as needed (setting up new lab space, consumables ordering, equipment maintenance) and generally being a good lab citizen.
  • Support and collaborate on multiple disease projects in a fast-paced startup environment, working across teams at ShapeTx.
  • Interpret and communicate results to team members to drive key decisions and future experiments.

Qualifications and Requirements

  • Bachelor's or Master's degree in a related discipline, with at least 2 years working in a laboratory setting.
  • Attention to detail, diligent record-keeping, organization, and time management.
  • An interest in technology development, learning new techniques, and inventing novel methods.

Skills and Experience

  • Experience with molecular biology techniques: RNA / DNA purification and sequencing; plasmid design and cloning; PCR.
  • Preferred experience in reverse transcription, library cloning, and Next Generation Sequencing.
  • Experience with mammalian cell culture and transfection, particularly cell line engineering using Crispr/Cas9, integrases, transposons, or viral transduction.
