Mammoth’s Discovery Team is seeking a Lead Scientist to establish and lead structural biology and engineering of CRISPR-associated (Cas) nucleases for therapeutic genome editing applications. Discovery is a cross-disciplinary team of researchers responsible for the identification, screening and optimization of novel Cas nucleases. The successful candidate will be positioned in a downstream role within the team and will be responsible for pushing Cas nucleases into pre-clinical applications. A strong background in structural biology (X-ray crystallography and/or cryo-EM) and structure-guided protein engineering is required for this role. 

  • Lead structural biology and rational protein engineering efforts at Mammoth
  • Manage external collaborations to support protein production and structural studies (CROs, academia)
  • Prepare protein, RNA, and/or DNA reagents and ensure quality specifications are met
  • Utilize computational and experimental approaches to design, produce, and evaluate engineered Cas nucleases for improved or altered activities
  • Document results and provide updates to key stakeholders

  • PhD in structural biology, molecular biology, synthetic biology or a related field
  • 6+ years experience designing, executing, and optimizing protein structural studies (X-ray crystallography and/or cryo-EM)
  • Demonstrated experience utilizing structure-informed engineering approaches to improve or alter the functionalities of proteins and/or RNAs (Eg. Rosetta, AlphaFold)
  • Computer and scripting skills (Benchling, Geneious, JMP, SQL, R, Python)

  • 3+ years biotechnology industry experience
  • Prior experience initiating and managing external collaborations in industry
  • Expertise in solving and analyzing the 3-D structures of nucleic-acid interacting proteins
  • Familiarity with CRISPR-Cas literature
  • Demonstrated ability to independently design and execute NGS assays
  • Hands-on experience with mammalian tissue culture and transfection
  • Ability to perform at a high level as a member of a cross-functional team
