Post-Doctoral Fellow, Platform Innovation
Synthorx and Ablynx, two companies now part of Sanofi, are seeking a postdoctoral researcher to work on a collaborative project. Synthorx has developed a “semi-synthetic organism” with an expanded genetic alphabet/code. The semi-synthetic organism stores an unnatural base pair in its DNA, transcribes it into RNA and then uses it to translate proteins with non-canonical amino acids specifically at encoded positions. Ablynx has pioneered the development of NANOBODY® VHHS, stable single-domain antibody fragments for therapeutic applications. The successful candidate will work collaboratively to combine the technologies – to use the semi-synthetic organism to produce Nanobodies with site-selectively incorporated bio-orthogonal handles that are used to couple multiple Nanobody building blocks and create bispecific constructs with novel architectures (to be used, for example, as T cell engagers). The successful candidate will also work towards optimizing the bio-orthogonal coupling and establishing screens to identify optimal architectures. The postdoc will be based at the Synthorx site in San Diego, California, but will also spend time at the Ablynx site in Ghent, Belgium. Publication will be a priority and on-the-job training will be provided.

Optimize the expression of different Nanobodies with non-canonical amino acids incorporated at different positions.
Optimize bio-orthogonal couplings.
Collaboratively evaluate Nanobody conjugates in vitro and in vivo using mouse models.
Explore the use of different non-canonical amino acids that enable different bio-orthogonal coupling chemistries.
Collaboratively work to develop assays to rapidly screen different conjugates.
PhD in appropriate field.
Experience with standard tools of molecular biology, including PCR, cloning, protein expression and purification, and/or bio-orthogonal coupling chemistry.
Must have permanent US work authorization.
