Over the past decade we have seen important advances in the cellular immunotherapy therapy field in particular, with dramatic rates of clinical response in hematological malignancies, and rapid evolution of our understanding of factors that could extend this success to solid tumors. However, these transformative therapies are often accompanied by severe and potentially life-threatening toxicities, and patient access is limited by such factors as complex, time-consuming patient-specific manufacturing, variable product quality, and cost.

Position Summary
Reporting to the Senior Director Discovery, the Senior Scientist NK Cell Immunotherapy will be responsible for generating and testing ideas for improving NK cell function to support the nomination of lead discovery candidate(s) for IND track development.

Role & Responsibilities

  • Design and develop CAR constructs.
  • Work with CROs to generate viral vectors, gene modify NK cells and characterize NK cell phenotype and function through well designed and executed proof-of-concept studies across NK cell discovery pipeline.
  • Identify and conceptualize new mechanisms to leverage NK cell biology and tumor microenvironment to enhance anti-tumor function.
  • Develop cell-based in vitro models and oversee animal models development in cross-functional teams or with CROs, to assess anti-tumor specificity, efficacy, and safety of gene-modified NK cells.
  • Gather, maintain, organize, and interpret experimental data in a timely manner.
  • Able to recognize problems and recommend/develop appropriate troubleshooting approaches.
  • Prepare and present findings to team members and cross-functional leadership teams.
  • Be proactive in cross-functional collaboration and training peers and scientific staff in experimental techniques as needed.
  • Maintain detailed records of experimental work in compliance with the company policy.

Experience, Education and Specialized Knowledge and Skills
Must thrive working in a fast-paced innovative environment while remaining flexible, proactive, resourceful and efficient. Ability to manage complex research and development functions and timelines through outsourced or partnered workflows is a requirement. Excellent interpersonal skills, ability to develop important relationships with key stakeholders, conflict management and negotiation skills, ability to analyze complex issues to develop relevant and realistic plans and recommendations. Demonstrated ability to translate strategy into action. Excellent analytical skills, ability to communicate complex issues in a simple way, and ability to orchestrate plans to resolve issues and mitigate risks.

  • Ph.D. in immunology, immune-oncology, tumor immunology (or a related field) with 5+ years of relevant experience in academic or biotech/pharma setting
  • Experience in cell culture, molecular biology techniques and in vitro techniques to assess NK cell functionality including but not limited to cytokine secretion, short and long-term cytotoxicity assays, NK cell activation assays, cell proliferation analysis.
  • Experience with high dimensional (multi-parameter) flow cytometry, flow antibody panel design and data analysis.
  • Experience in gene expression analysis is desired.
  • Prior experience in adoptive cell therapy and familiarity with techniques for genetically modified lymphocytes (CRISPR, Lentivirus etc.).
  • Proficiency in experimental techniques such as ELISA, MSD/Luminex, flow cytometry, viral transduction, electroporation, IncuCyte (or equivalent method); molecular methods such as chimeric gene design, sequence optimization, bi-cistronic gene expression design etc; as well as scientific software such as FlowJo, DNAStar Lasergene or equivalent, GraphPad Prism or equivalent.
  • Self-motivated and able to work on multiple projects as part of a team and as individual contributor, and prioritizing tasks to meet schedules and timelines.
