Pharmacokinetics & Drug Metabolism: Scientist

The department of pharmacokinetics and drug metabolism (PKDM) within Amgen Research is dedicated to the discovery and development of novel, effective, and safe medicines. PKDM is currently seeking a Scientist level candidate. The successful candidate will work in a team environment to characterize and refine the absorption, distribution, metabolism, excretion (ADME) and pharmacokinetic properties of therapeutic drug candidates to achieve their full pharmacological potential. PKDM works in close partnership with subject matter experts across many disciplines including medicinal chemistry, protein sciences, pharmacology, toxicology, pharmaceutical sciences, clinical pharmacology and regulatory to craft promising clinical therapeutic candidates, conduct preclinical development studies, support clinical evaluation and contribute to IND and NDA filings.

Job Description

The responsibilities of this position are to develop and employ bioanalytical, biochemical, biophysical and in vitro ADME assays to support portfolio and platform development of primarily small molecule drug entities. The successful candidate will design and execute LC-MS-based bioanalytical methods to quantify small molecules and endogenous proteins (i.e. biomarkers, protein targets, etc.) to support in vitro and in vivo ADME characterization and PK and PD analysis. The role may also include quantitation of oligonucleotides and protein therapeutics (i.e. antibodies, BiTE. etc.) in plasma, tissues, and other biological matrices. A background in small molecule drug metabolizing enzymes and/or transporters is strongly desired so that the candidate can also support DDI, TDI, MetID and phenotyping studies. This role will require data interpretation and communication of results to project team representatives in support of drug discovery and development and to support regulatory filing.

The ideal candidate will draw from any the following key technical skillsets:

  • Ability to design and execute experiments to quantitate molecular entities using LC-MS in support of both drug discovery and early development programs
  • Strong background in the use of biophysical, biochemical, and cell-based analytical methods to interrogate molecular attributes, including ADME properties and drug-target interactions
  • Primary hepatocyte and/or cell line culture experience is beneficial
  • Experience running assays to assess interactions with and/or activity of drug metabolizing enzymes is desired
  • Other responsibilities will include instrument maintenance and troubleshooting, electronic laboratory notebook keeping and communication of results to project teams.

Basic Qualifications

-Doctorate degree with 0-2 years of industry experience

Candidates for this PKDM Scientist position will have PhD or MS in chemistry, biochemistry, or equivalent. Experience with operation, troubleshooting, and data interpretation of LC-MS is required. Experience with operation of automated liquid sample handling instruments and immunoaffinity capture is a plus. The candidate should possess excellent oral and written communication skills and be able to excel in a highly collaborative, matrixed team environment.
