ROLE SUMMARY

The Cancer Immunology Discovery group at Pfizer is seeking an ambitious post-doctoral scientist with strong research and communication skills to undertake a highly visible research project within the T cell activation and PDx resistance team. The successful candidate will study macrophage and T cell interactions in the tumor microenvironment to identify spatial environmental cues that regulate the therapeutic response to immune checkpoint blockade. This work will extend research findings that we have published (Qu, Y., et al. (2020). Cell Rep 32(1)) and will address key concepts that we have also recently reviewed (Marcovecchio, P. M., et al. (2021). J Immunother Cancer 9(2)).

We are seeking a self-starting, highly motivated and technically proficient PhD level cellular immunologist to apply variety of tools including molecular, cellular and biochemical techniques alongside immunofluorescence imaging, high parameter flow cytometry and bioinformatics in preclinical in vivo models to address these key questions. (S)he will work in a fast-paced and highly collaborative research environment and will be expected to present their findings in high-impact journals and at both internal and external scientific conferences.

The overall aims of this project are to improve therapeutic outcomes for patients by improving our basic understanding of tumor-associated macrophage – T cell interactions in the tumor microenvironment. This work may inform the development of the next generation cancer immunotherapies and lead to improved patient selection strategies.

ROLE RESPONSIBILITIES

  • Design, conduct, analyze and interpret experiments to characterize temporal and spatial relationships between various T cell and myeloid populations within the tumor microenvironment.
  • Design and conduct adoptive transfer studies to determine the influence of T cell phenotype chemokine/chemokine ligand interactions on CD8 T cell localization and differentiation in response to anti-PD(L)1 treatment.
  • Determine cellular and molecular pathways that govern intratumoral myeloid differentiation.  
  • Design and conduct in vitro MoA studies to support in vivo genetic data

BASIC QUALIFICATIONS

  • PhD in immune-oncology or immunology
  • A high-level understanding of key concepts in cancer immunology and excellent working knowledge of either T cell or macrophage biology (preferably both).
  • Competent animal handling skills including intraperitoneal, sub cutaneous and tail vein injection.
  • Experience in ex vivo processing of tumor and lymphoid tissues
  • Expertise with primary cell isolation and culture assays, including co-culture and transwell assays
  • A strong desire to learn, it is not expected that the successful candidate will have experience with all of the required techniques however they must be willing to expand their technical and scientific capabilities

PREFERRED QUALIFICATIONS

  • Working knowledge of common murine syngeneic tumor models
  • Expertise with analysis of high dimensional datasets including single cell RNA-seq and multiparameter flow cytometry.
  • Experience performing high resolution immunofluorescence microscopy and data analysis
  • First author publications in high impact journals.
