TheBiochemistryand StructuralBiology(BSB)groupis seeking an accomplishedScientisttocontributetotheInduced-Proximity Platform(IPP)organizationtasked withthe discovery ofnoveltherapeuticsin theTargetedProteinDegradation field.

The BSBgroup is focused onenhancingourunderstanding of the biochemistry ofubiquitinligasesand other related enzymesinvolved in cellular protein degradation. These efforts including the discovery ofProteolysis Targeting Chimeras(PROTACs)andMolecular Gluesto advance Amgensdrug discoverypipelineofintractableand difficult-to-drugtherapeutic targets.

This role offers an enormousopportunityto conduct cutting-edgedrug discoveryresearchwithin a growing and science-driven organization. This is a uniqueopportunity for those whoare interested inconductingbasicresearch in the targeted protein degradation field while directlyimpacting therapeutic discovery programsat aworld-classorganization.

The candidate will work collaboratively with researchers across multiple functions withinthe IPPdepartment as well asacrossdiseasetherapeuticareasandtheTherapeuticsDiscoveryorganization.Thesuccessful candidatewillalsowork with external partners and contract researchorganizations(CROs)andhave the opportunity topresent workatnational and internationalconferences.

Key Responsibilities

  • Participate in the planning, design, execution, and documentation of laboratory studies related to the studying of protein complexes involved in proteindegradation.

  • Design ofDNAconstructs andtheuse of rational and computational tools for theengineeringofproteinconstructsto ensure successfulproteinexpression,purificationand structural determination.

  • Perform expression,purification,and characterization of recombinant proteins to support various aspects ofdrug discovery efforts related to Targeted ProteinDegradation.

  • Perform biochemical, biophysical and structural characterization of protein complexes.

  • Studyprotein-protein interactions.

  • Collaborate within and across interdisciplinary teams to advance ourunderstanding ofTargetedProteinDegradation mechanisms.

Basic Qualifications

  • PhDin Biochemistry, Protein Chemistry, Structural Biology, Chemistry or ChemicalEngineering.

  • Demonstrated scientificreasoning, dataanalysis and problem-solving skillsasevidencedby high impact scientific publications.

  • Highlymotivated and team player in across-functionalandfast-paced environment.

  • Work independently to plan, manage, and conductexperiments.

  • Ability to develop deep understanding of projects.

  • Strong understanding ofbasicprotein biochemistry and biophysicsprinciples, includingthose pertaining toprotein complex formation and protein transient interactions.

PreferredQualifications

  • 2 years of postdoctoralor industryexperience.

  • Prior experience with the purification and the biochemical and/or biophysicalcharacterization ofchallengingproteins andprotein complexes.

  • Expert user ofFPLC (AKTA PURE or Avant) and HPLC systems (WatersorAgilent).

  • Strong knowledge ofprotein analytical techniquesto studymacromoleculesincludingSEC, SEC-MALS, DLS, thermal shift assayand LC-MS.

  • Experiencein protein expression inE.coliand insect cells.

  • Practicalknowledge of techniques toinvestigateprotein interactions such as HDX, XL-MS, SPRand SAXS.Practical experienceinstructural biology including crystallography andcryoEMasdemonstrated by solvingmolecularstructuresofproteincomplexes.

  • Familiaritywithprotein modeling and design tools suchasMOE, RosettaorSchrdinger.

  • Familiaritywithstatisticaldata analysis tools such as JMPand R.

  • Knowledge of protein degradationmechanismsandin particularthebiochemistry ofubiquitinligases.
