4D Molecular Therapeutics, Inc. (“4DMT”) is a clinical-stage gene therapy company pioneering the development of product candidates using targeted and evolved AAV vectors. 4DMT seeks to unlock the full potential of gene therapy using its platform, Therapeutic Vector Evolution, which combines the power of directed evolution with approximately one billion synthetic capsid sequences to invent evolved vectors for use in targeted gene therapy products. The company is initially focused in three therapeutic areas: ophthalmology, cardiology, and pulmonology. A Sr. Scientist role in the Human Cell and Disease Modeling group at 4DMT will lead preclinical in vitro pharmacology work for AAV gene therapy candidates to treat a variety of diseases. The role allows for extensive creativity, innovation and opportunity to generate various cell systems and develop novel disease models to examine tropism, function and characteristics of AAV gene therapy vectors.  A Sr. Scientist works closely with HCDM team members and peers in other R&D departments to lead scientific programs, exchange ideas, improve protocols and generate valuable data to drive 4DMT towards its goals. They will participate in cross functional meetings and present data in various company settings. They are required to be scientifically independent and lead programs with little oversight.
  • Lab Activities: 50%
  • Stem cell culture maintenance and reprogramming
  • Optimization of cell differentiation protocols in 2D and 3D culture systems
  • Ability to effectively design experiments to characterize cell models and determine tropism of AAV capsids
  • Development of functional assays to determine efficacy of lead AAV vectors
  • Draft study designs, protocols, differentiation paradigms and maintain an electronic lab notebook
  • Design directed experiments to meet strict timelines
  • Cross Functional/Communication: 30%
  • Contribute scientific ideas and knowledge in departmental and cross-functional team meetings
  • Effectively present findings at departmental and cross-functional team meetings
  • Engage in HCDM study design for various programs
  • Present annually at Research and Progress meeting
  • Help compile and write data sections for IND preclinical pharmacology reports
  • Communicate effectively with management and their team to achieve departmental goals

  • Ph.D. in a relevant scientific field, preferably Cell and Molecular Biology
  • Experience:
  • 10+ years of experience within Biotech or similar industry
  • 8+ years of cell culture experience
  • 5+ years of stem cell culture maintenance and reprogramming experience
  • 2+ years management experience in Biotech or similar industry
  • Technical Skills
  • Expertise in cell culture: adherent, suspension, monolayer, 3D
  • Expertise in stem cell culture, maintenance, freezing and passaging
  • Fibroblast reprogramming into pluripotent stem cells through various non-integrating methods
  • Ability to develop and carry out differentiation of stem cells into various cell types
  • Protein identification through Western blotting, flow cytometry and immunocytochemistry
  • Development and optimization of functional assays for various cell types
  • Expertise in microscopy and using imaging software
  • Strong in molecular biology techniques and virus handling and transduction

  • Physical Requirements
  • Lifting no more than 20 lbs to help put away supplies
  • Ability to sit and work at a cell culture laminar flow hood and at a microscope for several hours at a time
  • During COVID-19, this person needs to be in a lab at least four times a week and adhere to 4DMT Covid protocols and policy
