Job Details
characterize them by performing various in vitro and in vivo assays to measure their infectivity and ability to transduce target cells of interest. Projects will
involve using various mouse models and a variety of assays to monitor transduction, such as bioluminescent imaging, immunohistochemistry, in situ hybridization, vector genome quantification from tissue, flow cytometry, and ELISA.
As an R&D Specialist in Viral Vector Technologies, a typical day may include the following:
- Maintain, transfect, and transduce mammalian cell cultures
- Produce, purify, quantify, and characterize viral vectors in BLS2 cell culture for in vivo studies
- Dose mice with viral vectors and/or other therapies via tailvein (IV) injection, IP, IM, or SubQ routes
- Monitor infection/biodistribution of viral vectors through in vivo bioluminescent imaging, ex vivo fluorescence imaging, immunohistochemistry, in situ
hybridization or by blood draws and serum/plasma collections
- Collect biological samples from mice through dissections; process samples appropriately for downstream analysis (immunohistochemistry, qPCR, RNAScope/BaseScope, flow cytometry)
- Assess non-human primate biological samples (PBMCs, plasma/serum, FFPE tissue)
- Perform basic biochemical/immunological assays (ELISA, flow cytometry, Luminex/MSD)
- Perform basic molecular biology assays (Cloning, Western blotting, DNA/RNA extractions, PCR, RT-PCR, quantitative PCR, etc.)
- Analyze and present data (familitarity with Graphpad Prism or other statistical/graphical software)
- Maintain lab supplies and help with lab organization
This role may be for you if:

- You are able to handle multiple projects and are comfortable changing gears quickly as needed
To be considered for this role, you must have a BS/MS with 3 + years of experience. Experience with animal handling, animal injections (specifically tailvein), bleeds (retro-orbital, tail-nick, etc.) is required. Experience with serum/plasma collection, bioluminescent imaging, mouse dissections and tissue analysis by various methods is required. Experience with mammalian cell culture, molecular biology, and biochemistry techniques is preferred. Experience working with Adeno-associated virus (AAV) or CRISPR/Cas9 is a plus.
