Adverum Biotechnologies is a gene therapy company committed to discovering and developing novel medicines that can offer life-changing benefits to patients living with rare diseases or diseases of the eye who currently have limited or burdensome treatment options. Adverum has a robust pipeline and is leveraging its next generation adeno-associated virus (AAV)-based directed evolution platform to generate product candidates designed to provide durable efficacy by inducing sustained expression of a therapeutic protein. Our focus on the patient is supported by clinical development expertise and core capabilities in vector optimization, process development, manufacturing, and assay development.
 
This is a research position for a highly motivated bench scientist to support the Research and New Vector Technologies group. The candidate will support the early development and evaluation of AAV-based therapeutics, including vector optimization, development of in vitro cell system, and the development of research-grade assay in order to conduct in vivo biodistribution and pharmacokinetic studies.
 
This is a contract to hire role (conversion within a minimum of 3 months based on performance).
What you'll do:
  • Spend 80% their time performing bench-level research
  • Independently plan and execute experiments, analyze and communicate results.
  • Actively apply new concepts and technologies. Be proactive in solving technical problems through analyzing data and troubleshooting methods.
  • Perform a broad range of cell and molecular biology techniques (e.g. DNA and RNA isolation, cDNA synthesis, qPCR, immunoblotting, immunofluorescence, flow cytometry, etc.)
  • Provide bionanaytical support to in vivo preclinical studies to quantify AAV vector pharmacokinetics and biodistribution, and therapeutic rescue/safety, using molecular and histological analyses.
  • Develop research-grade assays (WB, ELISA, MSD, qPCR, RTqPCR, immunohistochemistry).
  • Participate in the design, cloning and optimization of novel AAV vector constructs (with and without collaboration with external CROs). This also includes in vitro screening for expression and functionality, in cell lines and tissue explant cultures. In vitro evaluation and screening of new AAV variants.
  • Support analysis of AAV vector lots, including titration, WB, SS, endotoxin assay, gel analysis and support of NGS analysis in coordination with CRO.
  • Support and contribute to the development of AAV in vitro expression and functional assays. This includes the screening of preexisting cell lines or genetic engineering to generate stable cell lines.
  • Initiate and manage external research collaborations with contract research organizations and academic investigators in a diverse array of scientific disciplines.
  • Supervise and train technical staff.
  • Support and assist the members of the team, as needed.

About you:
  • Minimum 8 years (MS) or 3 years (PhD) experience in gene delivery/therapy, bioengineering, biochemistry, biotechnology, virology or related (industry or academic setting).
  • Solid proficiency and demonstrated track record in molecular biology including but not limited to molecular cloning, site-directed mutagenesis, PCR, mRNA and DNA quantification/analysis (Southern Blot, Taqman primer design and validation, RT/qPCR, ddPCR), sequencing (Sanger, NGS and bioinformatic analysis), protein analysis and immunoassays (Western Blot, ELISA, MSD),plate-based reporter and/or enzymatic activity assays.
  • Previous experience with monoclonal antibody screens, characterization and immunoassay development/optimization is preferred.
  • Experience in immunocytochemistry/immunohistochemistry procedures is a plus.
  • Strong cell culture skills with a demonstrated track record: sterile technique for handling and maintenance of mammalian cells, non-viral gene transfer and editing (transfection, electroporation) gene editing (CRISPR or TALEN), viral infection (AAV, Lentivirus and/or Adenovirus), flow cytometry and cell-based assay development. Solid experience with cell engineering and cell line generation is preferred.
  • Experience with tissue explant cultures, primary cell lines and/or iPS is a plus.
  • Demonstrated ability to work independently or in a team-oriented environment with moderate supervision.
  • Excellent data analysis skills and experience with a variety of scientific software applications is required (including Microsoft Office, Prism, Geneious, Adobe Photoshop, Illustrator).
  • Strong experimental design and demonstrable track record of solving technical problems through analyzing data and troubleshooting method issues.
  • Excellent communication, presentation, collaboration, time-management, and organization skills are required.
  • Proven ability to work independently in hands-on laboratory setting, self-motivated to learn and develop new techniques, perform multiple tasks simultaneously, keep accurate and timely records, follow instructions, and comply with company policies.
  • The successful candidate will be an ambitious self-starter, be motivated to apply his/her skills to diverse projects and enjoy working in a fast-paced team environment. Familiarity with drug product development, creating SOPs and GxP is a plus.
  • Work authorization in the US required.
