Harpoon Therapeutics is a clinical-stage immunotherapy company developing a novel class of T cell engagers that harness the power of the body’s immune system to treat patients suffering from cancer and other diseases. T cell engagers are engineered proteins that direct a patient’s own T cells to kill target cells that express specific proteins, or antigens, carried by the target cells. Using its proprietary Tri-specific T cell Activating Construct (TriTAC®) platform, Harpoon is developing a pipeline of novel TriTACs initially focused on the treatment of solid tumors and hematologic malignancies. HPN424 targets PSMA and is in a Phase 1/2a trial for metastatic castration-resistant prostate cancer. HPN536 targets mesothelin and is in a Phase 1/2a trial for cancers expressing mesothelin, initially focused on ovarian and pancreatic cancers. HPN217 targets BCMA and is in a Phase 1/2 trial for relapsed, refractory multiple myeloma. HPN328 targets DLL3 and is in a Phase 1/2 trial for small cell lung cancer and other DLL3-associated tumors. Harpoon has also developed a proprietary ProTriTAC™ platform, which applies a prodrug concept to its TriTAC platform to create a therapeutic T cell engager that remains inactive until it reaches the tumor. We are currently seeking a Research Associate to join the New Technologies Group at Harpoon. Our growing team is focused on the development of novel, conditionally active biologic prodrugs, including our protease-activated T cell engager platform (ProTriTAC). These prodrugs are designed to be preferentially active in the tumor, and thus enable the safe targeting of previously intractable therapeutic targets. The successful candidate will be expected to generate in vivo proof-of-concept data for Harpoon’s ProTriTAC drug candidates, work collaboratively in a team-oriented environment, and have a strong desire to learn new techniques and their underlying scientific rationales.

KEY RESPONSIBILITIES

  • Design and execute in vivo proof-of-concept experiment using mouse tumor models and PK studies
  • Perform mouse tumor models from start to finish: expansion of tumor cell lines and immune cells in vitro, enumeration of cell density, implantation of tumor and immune cells in vivo, measurement of clinical endpoints, and analysis of data
  • Maintain cultures of tumor cell lines and immune cells
  • Prepare test articles with appropriate serial dilutions
  • Dose mice via multiple routes of administration
  • Perform blood collection, tissue extraction, and tissue processing
  • Perform in vitro biochemical and cell-based assays to support in vivo data findings
  • Collect, analyze, and present experimental data regularly at team meetings
  • Maintain clear and complete experimental records with an electronic lab notebook
  • Support project needs as they arise

MINIMUM QUALIFICATIONS

  • BS or MS in biology or a related discipline with 2+ years of laboratory experience
  • Proficiency with mouse in vivo handling skills, including administration of test articles by sc, ip and iv injections, implantation of tumor and/or immune cells, and collection of blood and tissues
  • Meticulous experimentalist with strong attention to detail
  • Effective verbal and written communication
  • Ability to work independently with limited supervision
  • Ability to accommodate a flexible schedule, which may include occasional weekend responsibilities

PREFERRED QUALIFICATIONS

  • Direct hands-on experience and working knowledge of in vivo mouse tumor models, including xenograft and/or syngeneic tumor models, and mouse PK studies
  • Ability to design and execute multi-color flow cytometry experiments and analyze data with FlowJo, FACSDiva, or similar analysis programs
