Senti Bio is seeking a collaborative and passionate Scientist to join our Gene Circuit Discovery group. This is an individual contributor role that will apply expertise in synthetic biology to design and test gene circuits for the generation of regulated cell and gene therapies in primary immune cells (e.g. NK cells, T cells, macrophages, and/or monocytes). The ideal candidate will be comfortable planning and independently executing in vitro experiments in immune cells, communicating findings, and collaborating with Senti colleagues to push projects forward in a fast-paced startup environment.     • Design and optimize therapeutic genetic circuits composed of parts including engineered proteins, synthetic promoters, and RNA-based regulatory elements
    • Generate viral vectors for gene circuit transduction into multiple mammalian cell types including primary immune cells
    • Evaluate gene circuit performance using a variety of in vitro assays including multi-color flow cytometry, microscopy, ELISA, and Luminex
    • Analyze data and present results to downstream stakeholders
    • Document experiments by generating timely records of protocols and data using Senti's electronic lab notebook
    • Collaborate with your colleagues at Senti by coordinating with other group members and research teams to troubleshoot problems, develop new assays, and support cross-functional projects
    • Stay current with the relevant scientific literature, and participate in ideation and decision-making meetings
    • Provide expert support for experiment design and assay development to the broader scientific team

    • Ph.D. or equivalent with expertise in synthetic biology, immunoengineering, bioengineering, biotechnology, or cell engineering
    • Proficient with mammalian cell culture and mammalian cell engineering, preferably for therapeutic applications
    • Capable of designing and implementing multicolor flow cytometry experiments
    • Desire to be a team player in a fast-paced environment
    • Comfortable presenting work to cross-functional groups

Preferred Qualifications
    • Proficient in primary immune cell culture and engineering by lentiviral or other methods (e.g. NK cells, T cells, macrophages, and/or monocytes)
    • Experience with assaying immune cell states and behaviors, including cell surface markers, cytokines, migration, and cytotoxicity
    • Experience with NGS sample prep and analysis
    • Molecular understanding of transcriptional regulation
    • Familiarity with FlowJo, SnapGene, (Prism and Benchling are also a plus)
    • Experience in mammalian synthetic biology is a plus, such as design of synthetic transcriptional and/or translational regulation to control therapeutic gene expression
