These positions are in ES Cell Culture within Velocigene in Tarrytown, NY. There are 2 openings for Summer 2020. Each opening is described below.

Position 1:
Cre/Dre/Flpo optimization of rESC nucleofection

This student will complete a project that will include the comparison of plasmid, mRNA and protein of Cre, Dre and Flpo with the read-out using digital PCR and/or cell counter (Cellometer). The student will observe and help with the culture and nucleofection of rESC. During this time, we would continuously culture rESC and perform EPs weekly. After nucleofection, the EP'd cells will be plated onto laminin to be processed by the student. The student will harvest the cells, extract DNA and perform digital PCR under supervision with the goal to identify the best conditions for Cre, Dre and Flpo EPs in rESC.

Position 2:
The Embryonic Stem Cell Lab at Velocigene is looking for a summer intern to assist with quantitative image analysis projects. This internship will focus on the extraction of information from microscopy images and will consist of very limited wet lab work. The summer intern will be working primarily with Nikon's software platform, "NIS-Elements" to build invitro imaging analysis tools.

Sarcomeric function
Human cardiomyopathies are characterized by impaired diastolic and systolic performance. These parameters are difficult to assess in rodent models. Instead we propose an invitro system of analyzing contractility and relaxation of patient derived induced Pluripotent Stem Cell Cardiomyocytes (hiPS-CM) to model biophysical functions of sarcomeres. Image based analysis of sarcomere lengths in cardiomyocytes across the contractile cycle would allow for quantitative proxies of systolic (percent shortening) and diastolic function (duration of relaxation).
This human invitro cardiac project would have the intern generating end point readouts using image based quantitative measurements of sarcomeric lengths of hiPS-CM. -cellular shortening (%) -relaxed and contracted sarcomeric lengths (um), contractile cycles, and relaxation durations (seconds).

Barrier integrity assay
An invitro neurovascular unit is a model of the human blood brain barrier (BBB) which consists of neuronal cell types and endothelial cells. Barrier function of the BBB model can be assessed using a real-time barrier integrity assay. In this assay, a fluorescent dye is perfused through the lumen of the model's endothelial vessel. In case of a leak-tight barrier, all dye is retained in the microvessel, while in case of a leaky barrier or a cell-free control, the fluorescent dye leaks into the adjacent ECM gel channel. The fluorescent intensity in both channels is monitored over time and is used to quantify barrier function in each condition.

Functional Transport Assay
A different assay can be employed to assess the function of one of the BBB's main efflux transporters, the P-glycoprotein (P-gp) transporter. Calcein-AM can be perfused through the lumen of the BBB's endothelial vessel and taken up by the cells, making the cells green-fluorescent. In the presence of a P-gp inhibitor, P-gp efflux of calcein is reduced, resulting in accumulation of green-fluorescent signal inside the cells. The endothelial cells forming the model's barrier can be imaged and the green-fluorescent signal per cell calculated using an additional Hoechst staining to visualize the nuclei. In presence of a P-gp inhibitor, the endothelial cells of the BBB model should show increased green fluorescence, indicative of functional P-gp transport.

This invitro neurovascular project would have the intern generating end point readouts using image based quantitative measurements of fluorescence.

Position 1:

• Must be pursuing a Bachelor's Degree

• Must be pursuing a degree in Biology, Biotechnology, Molecular Biology, Biomedical Sciences, Biochemistry, or related discipline

• Desired technical skills include, but are not limited to, experience in cell culture, experience in molecular biology particularly DNA and RNA analysis and PCR, and good organization and communication skills. Dedication to team work and process oriented are essential

Position 2:

• Must be pursuing a Bachelor's or Master's Degree

• Must be pursuing a degree in Electrical Engineering, Computer Science, Biological Sciences or related discipline

• Must have experience with image analysis, computer literacy, and coding experience

General Intern Program Information:

• Must be enrolled in, or accepted to, an academic program pursuing a Bachelor's, Master's, PhD or PharmD

• Summer Program is full-time (~40 hrs/wk) for at least 10 weeks and is paid

• Prefer demonstrated leadership in areas such as campus activities, clubs, sports or the community

• Minimum GPA of 3.0

• You will work with a specific manager on a project(s)

• Enjoy weekly 1 hour Intern Program events: learn about other areas of the company from VPs and other employees, soft skills workshops, networking, volunteering, and more!

• Present to your team on your summer project
