Summary:

The successful candidate will work in close collaboration with the lead microcopy scientist to develop, optimize, automate, and execute new histology, immunohistochemistry and in situ hybridization protocols for preclinical rodent model studies throughout all therapeutic focus areas of Research. Together with the lead microscopy scientist, this role will also coordinate histotechnological protocols, instrumentation, techniques, and reagents throughout Research, including training and support of scientific personnel. Regeneron's broad and ever evolving preclinical research portfolio will require the candidate to be independent, organized, and able to coordinate work flow and deliverables with the internal and extended teams, all while maintaining a high standard of quality.

Current and emerging projects include: optimization of IHC/IF protocols to detect in vivo administration of Regeneron antibodies across a variety of tissue types, characterization of targets in lung fibrosis, detection and characterization of peripheral nerve enervation in various mouse models, and optimization of multiplexec IHC/ISH fluorescence protocols to support spectral unmixing imaging for immune cell profiling in tumor microenvironment. Projects are constantly evolving and will change as new ideas and opportunities present themselves.

Responsibilities:

• Working closely with the lead microscopy scientist to understand project needs and priorities, troubleshoot experiments, and provide updates on the progress of projects.

• Independent execution of projects planned in consultation with the lead microcopy scientist and other collaborating scientists.

• Development of histological techniques including antibody and probe validation, optimization across multiple tissue types, combined IHC/ISH protocols, quenching autofluorescence, multiplexed fluorescence RNAScope, and more

• Handling small animals including euthanasia followed by intracardial perfusion and dissection for tissue harvesting.

• Experimental animal tissue dissection, sample preparation, sectioning, staining, and histological analysis of multiple animal models.

• Testing, integration, and support of new robotics technologies and assays to streamline and automate histological protocols.

• Lab operations including organization of materials and reagents required for studies and maintainance of histology equipment to ensure high quality laboratory practices are followed.

• Maintain online and physical records of the data from ongoing and completed projects.

Requirements:

8 years lab experience

The position requires a Bachelors degree in science having at least 8 years of relevant experience.

• Extensive experience in basic and advanced histological techniques such as cryo and paraffin sectioning, immunohistochemistry, immunofluorescence, and in situ hybridization, including RNAScope and multiplexed technologies. Experience with resin/plastic embedding and sectioning is a bonus.

• Experience with histological robotic equipment such as the Leica BOND Rx or Ventana Discovery Ultra, and with white light and fluorescence digital pathology slide scanners such as the Aperio AT2, Leica SCN400, Zeiss Axioscan, or similar.

• Substantial knowledge of vertebrate anatomy

• A proven history of executing and accomplishing projects in a timely and independent manner within a laboratory setting is required, pharmaceutical industry experience is preferred.
