Summary:
Regeneron is currently seeking a talented and motivated Associate Scientist / Scientist to join a technology development group which aims to develop novel viral vectors. The candidate will develop novel viral vectors designed to target specific tissues and cell types, and will explore and modulate the innate and adaptive immune responses during Adeno-associated virus (AAV)-based gene therapy. The successful candidate should have a strong background in immunology. Experience with in vivo mouse models and flow cytometry is required. Experience in gene therapy, specifically producing or working with AAV, is a plus. Incumbent should be able to work methodically and with minimal supervision. The candidate will design and execute experiments to characterize novel viral vectors in vitro and in vivo. Experience with tissue culture and handling cell lines is required. Specific experience should ideally include immunological assays such as ELISA, ELISpot and flow cytometry, basic molecular biology, cloning, mammalian cell culture, mouse handling, and manipulation and purification of viruses in BSL2 conditions.

Responsibilities:
1. Maintain, transfect, and transduce mammalian cells in culture
2. Produce, purify, quantify, and characterize viral vectors
3. Perform basic molecular biology assays (Cloning, Western blotting, DNA/RNA extractions, PCR, RT-PCR, quantitative PCR, etc.)
4. Perform basic biochemical/immunological assays (ELISA, flow cytometry, ELISpot)
5. Dose mice with viral vectors and/or other treatments via tailvein injection, IP, IM, or SubQ route
6. Monitor infection through in vivo bioluminescent imaging or by blood draws and serum collections
7. Collect biological samples from infected mice through dissections and blood draws; process samples appropriately for downstream analysis (multiparameter flow cytometry, immunohistochemistry, qPCR, RNAScope)
8. Manipulate viral vectors in BSL2 cell culture
9. Analyze and present data
10. Maintain lab supplies and help with lab organization
Requirements:
TECHNICAL SKILLS:
Cell culture, transfections, AAV production and purification, virology, basic cellular and molecular biology/biochemistry, flow cytometry, virus infections, animal handling and injections (specifically tailvein), bleeds and serum collection, dissection techniques, ELISAs, ELISpot analysis

CORE COMPETENCIES:
- Incumbent should be skilled in designing well-controlled experiments that can provide reliable results
- A deep understanding of immunology is required
- Incumbent must have experience in working with mice, specifically injecting compounds via various routes of injection (tailvein is important)
- Incumbent must have experience with various technique used to characterize immune responses (ELISA, ELISpot, multi-parameter flow cytometry)
- Experience with mouse dissections and tissue analysis by various methods is desired
- Incumbent should have experience with basic safety procedures and sterile techniques required for BSL2 laboratory function
- Incumbent should have experience in molecular biology, specifically cloning and western blotting
- Understanding and experience with in vitro assays using viral vectors will be a distinct advantage
- Incumbent should be able to analyze data and extract conclusions
