Job Details

Known for its scientific and operational excellence, Regeneron is a leading science-based biopharmaceutical company that discovers, invents, develops, manufactures, and commercializes medicines for the treatment of serious medical conditions. Regeneron commercializes medicines for eye diseases, high LDL-cholesterol, atopic dermatitis and a rare inflammatory condition and has product candidates in development in other areas of high unmet medical need, including rheumatoid arthritis, asthma, pain, cancer and infectious diseases.

These positions are in Formulation Development in Tarrytown, NY. There are 4 openings for Spring AND Summer (Full time, about 7 months from January to August 2020). Each position is described below.

Position 1:
The student will work in a state-of-the-art laboratory under mentorship of a Scientist to support the development of biologic drug products which may include therapeutic antibodies, bispecific antibody and AAV based gene therapy. The student will support the daily activities of analytical development, stability testing and formulation development. The co-op student will have opportunities to learn fundamentals of drug product development, Design of Experiment and work on the cutting-edge technologies.
The main responsibilities include:
1. Gain experience in conducting formulation screening and stability studies
2. Gain hands-on experience in using UPLC and other analytical instruments to support formulation development
3. Gain experience in product stability data analysis by assisting database building and large data analysis using statistic approaches
4. Assist with operations and maintenance of UPLC systems
5. Assist in routine buffer and reagent preparation
6. Document the experiments and data according to the department policy
7. Present research findings at the group and departmental meetings

Position 2:
This student will investigate the underlying mechanisms by which proteins self-associate and ultimately form large transient structures in solution. A combination of molecular modeling approaches and biophysical approaches will be used to study the effect of various exipients on strength of protein-protein interactions, viscoelasiticity of formulated solutions and ultimately protein structure. Student will collaborate with researchers across departments to harness subject matter expertise and develop a holistic understanding of protein-excipient interactions that ultimately manifest into favorable solution properties for biologics formulations.

Position 3:
This student will develop platform models for freeze-thaw and mixing unit operations, important processes during the drug product fill finish manufacturing. Different factors such as protein concentration, excipient compositions, protein types will be investigated for validating the models. The outcome of these studies will provide deep understanding and recommendation on freeze-thaw and mixing operation parameters to manufacturing site. The student will also participate in other drug product process development studies such as filtration, shear characterization, and filling in program support for late stage development. The student will gain basic knowledge and experience in the drug product fill finish process development and manufacturing.

Position 4:
This student will conduct experiments to understand and assess degradation pathways related to a fusion protein being developed for ophthalmology indications. Aggregation is a common degradation pathway for proteins. It is important to understand the impact of aggregation on the efficacy and quality of therapeutic protein drug products. Experiments will be performed to understand the aggregate formation as a function of various factors such as time, temperature, protein concentration, stabilizer level etc. Reversibility or dissociability of aggregates formed under stress and storage conditions will be assessed using a variety of biophysical and analytical techniques such as Dynamic Light Scattering, HPLC etc. Experiments will also be performed in the presence of in vivo fluids such as serum etc. to understand the aggregation behavior of protein under physiological conditions. Lastly, impact of other product attributes such as viscosity will also be assessed during these experiments.
