Job Details

The Regeneron Genetics Center (RGC) is a wholly-owned subsidiary of the Company, whose goals are to apply large scale human genetics to identify new drug targets and to guide the development of therapeutics programs and precision medicine. Building upon Regeneron's strengths in mouse genetics and genetics-driven drug discovery and development, the RGC specializes in ultra-high-throughput exome sequencing, large scale informatics and data analysis encompassing genomics and electronic health records, and translating genetic discoveries into new biology and drug discovery opportunities. The RGC leverages multiple approaches including large population based studies, Mendelian genetics and family based studies, founder population genetics, and large-scale disease focused projects and has developed a network of over 50 collaborations with research organizations around the world. Including some of the largest sequencing studies in the world, such as the DiscovEHR study in collaboration with Geisinger Health System, and an initiative to sequence 500,000 participants with the UK Biobank, the RGC has built one of the largest human genetics databases, including sequence data from over several hundred thousand participants and rapidly growing. Our interests encompass a breadth of different areas across all therapeutic areas and the RGC is highly integrated into all facets of research and development at Regeneron. Program goals include target discovery, indication discovery, and patient-disease stratification. Objectives include advancing basic science around the world through public sharing of discoveries, providing clinically-valuable insights to physicians and providers of collaborating health-care systems, improving patient outcomes, and identifying novel targets for drug development.

Summary:
The overall goals of this position are to conduct analyses in founder and other unique populations, interpret genetic data in order to obtain a deeper understanding of human biology, bioinformatically support analysis and identification of novel targets, and seek indications for drug programs. The successful candidate will work as part of the Founder & Special Populations team as well collaborate with the RGC Analytical Genetics and Therapeutic Area Genetics teams to provide genetic, informatics, and biological support for genetics studies of founder and special populations. The successful candidate will: characterize the allelic architecture of many founder populations, provide bioinformatics support for family-based analyses, identify genes of potential interest through genotype-phenotype associations & other genetic approaches, harness internal & external data sources to better understand novel genetic findings, work with the Analytical Genetics group to seek validation of associations in other RGC population resources, and work with the Translational Genetics & Functional Modeling groups to glean deeper insights into mechanisms & biology.

Responsibilities:

• Ability to work collaboratively as a member of the Founder & Special Populations team, with other Regeneron teams, and with external collaborators

• Manage multiple projects simultaneously and complete analyses in a timely & reliable manner

• Catalog and interpret results of allelic architecture analyses in various founder populations to glean novel insights into human biology and develop follow-up strategies

• Interpret results of genotype:phenotype association analyses from whole exome sequencing and genotyping studies

• Apply biological insights, internal and external data sets, published literature, and other resources to prioritize genetic discoveries in founder and other unique populations for functional follow-up

• Provide bioinformatics support for the Mendelian collaborations

• Work with Analytical Genetics and Therapeutic Area Genetics teams to validate potentially interesting findings in founder and other special populations by extending the analyses to other RGC study populations (ex. Geisinger DiscovEHR, UK Biobank)

• Work with Functional Modeling, Therapeutic Focus Area, and Early Clinical Development teams to nominate, design call-back studies, and functional experiments to follow-up on interesting hits

• Summarize and communicate study findings to the Founder and Special Populations team, leadership, and internal and external collaborators

• Survey the literature and attend conferences to incorporate discoveries and findings from the broader genetics community into study opportunities and considerations internally
Requirements:

• This position requires a PhD with 2+ years of experience with a strong background in human disease and/or applied population genetics

• Some experience analyzing and interpreting whole exome sequencing and genotyping studies (ex. variant level QC, statistical association analysis, and interpretation of association signals in complex disease)

• Some experience with standard statistical approaches and tools (ex. regression, linear mixed models, PLINK, R) and some programming experience (ex. Linux, Python)

• Ability to summarize data and results to identify patterns, signals, and issues

• An understanding of biology to integrate and leverage diverse datasets to identify interesting variants, genes, and genotype-phenotype associations

• Experience with molecular characterization and functional studies geared towards elucidating gene functions and the mechanistic basis for genetic associations is a plus
