The Amgen Postdoctoral Program is committed to providing future scientists with an enriched environment to inspire innovation that will contribute to the development of human therapeutics.

Attribute Sciences organization within Amgen Process Development is seeking a Postdoctoral Fellow in the area of characterization of posttranslational and other chemical modifications of therapeutic proteins impacting their potency and life duration in human circulation. Attribute Sciences play critical role in development and commercialization of protein drug candidates and development of novel analytical techniques. The Postdoctoral Fellow will work under supervision of Dr. Pavel V Bondarenko. Manuscript publication and meeting presentations will occur with the outcomes of the studies.

Identify, characterize attributes (post-translational and other chemical modifications) of therapeutic proteins and establish their biological impact on efficacy and pharmacokinetics.

The chemical modifications of interest include clipping, glycosylation, oxidation, deamidation, aspartate isomerization, hydroxylation, fragmentation and multiple other modifications

Develop and apply novel sample preparation and analytical methods with emphasis on chromatography and mass spectrometry (LC-MS/MS) for characterization of intact, reduced therapeutic proteins and after enzymatic digestion by peptide mapping.

Work in an analytical group equipped with several Agilent HPLC systems and mass spectrometers including Thermo Orbitrap, Q-Exactive, Waters Q-TOF

Communicate verbally and in writing scientific concepts and results

Create publications and patents

Work across organization with other scientists on multidisciplinary teams

Follow protocols and procedures, safety requirements, maintain accurate records in an electronic lab notebook, and generate quality data for use in novel method development, therapeutic protein characterization

The assignment length is expected to be 3 years long.

Basic Qualifications

PhD at the time of appointment

Preferred Qualifications

PhD in Analytical Chemistry, Biochemistry

Documented theoretical knowledge and hand-on experience in peptide mapping of proteins, intact and reduced LC-MS analysis for identification and quantitation of chemical modifications

Training and experience of operating Thermo Orbitrap, Q-Exactive, Waters Q-TOF mass spectrometers, Agilent HPLC

Experience with affinity purification, including immobilizing target proteins on resin and utilizing it to purify the proteins of interest from complex biological matrixes
