Precision Biosciences is seeking a Cell/Molecular Biologist to help characterize the activity of ARCUS nucleases in cell-based assays utilizing primary cell types derived from human and non-human tissues. The Scientist will collaborate with various Precision Biosciences teams to develop phenotypic assays that enable translation of nuclease activity to biologically relevant outcomes. The early elaboration of this pharmacological information is an important step in the progression of ARCUS-based therapeutics from discovery to pre-clinical development.

  • Characterize ARCUS-modified cells at the molecular and cellular level.
    • Deliver ARCUS Nucleases into cultured transformed and primary cells and assess the phenotypic response of ARCUS-modified cells
    • Analyze cell-based experiments at the molecular level (DNA, RNA or protein) via indel assays, fluorescent analysis by FACS, PCR/digital PCR or ELISA
  • Utilize ARCUS Nucleases to generate proof-of-concept data demonstrating the validity of the therapeutic approach or mode of action in model system(s)
  • Identify or create appropriate cell lines/models for a given target/approach
    • Characterize cell line/model.
    • Create cell lines and/or reporter assays
  • Provide Molecular support for the team:
    • Construct specialized plasmid-based DNA clones for use in all ongoing projects
    • Provide high quality DNA stocks and sequencing analysis
    • Characterize ARCUS Nucleases for a given target
      • Optimize delivery mode (DNA, RNA, protein)
  • Test refined ARCUS Nucleases to improve function
  • Cultivate mammalian cells
    • Maintain CHO KI, HEK 293, and other laboratory cell lines and frozen stocks
    • Create, characterize, and maintain novel reporter cell lines
    • Transfect and transduce mammalian cells
  • Maintain accurate records in ELN system and assist in the organization and tracking of lab constructs, cell lines, and supplies
    • Contribute to the generation of SOPs in support of pre-clinical data information
  • Assist in general operation of the cell culture and molecular lab
    • Monitoring/changing CO2 tanks
    • Monitoring LN2 storage tank
    • Ordering and stocking of lab consumables
    • Disposal of Waste
  • Report results to Therapeutic Discovery Team
    • File reports for internal use and for partners
    • Generate graphs, slides, protocols and other documents for partners

The requirements listed below are representative of the knowledge, skill, and/or ability required to perform this job successfully.


  • The Scientist should have a PhD with 2-5 years of relevant experience or equivalent training and experience in the following areas:
    • Cell Culture
    • Primary cell propagation in culture
    • Cell Based Assay development
    • Plasmid and genomic DNA isolation
    • RNA isolation
    • Molecular Cloning techniques for Vector construction
    • PCR, qPCR
    • DNA Sequencing
    • DNA, RNA Transfection and Viral Transduction for expression in Eukaryotic Cells Systems
    • Development, characterization and optimization of bioanalytical methods for gene expression and protein analysis
    • Protein Characterization by Western, Flow Cytometry, and ELISA
  • Experience with genome editing is strongly preferred

Language Ability
  • Ability to read, analyze, and interpret common scientific and technical journals, financial reports, and legal documents
  • Ability to respond to common inquiries or concerns from business partners, regulatory agencies, or members of the business community

Math Ability
  • Ability to work with mathematical concepts such as probability and statistical inference, and fundamentals of plane and solid geometry and trigonometry
  • Ability to apply concepts such as fractions, percentages, ratios, and proportions to practical situations

Reasoning Ability
  • Ability to define problems, collect data, establish facts, and draw valid conclusions
  • Ability to interpret an extensive variety of technical instructions in mathematical or diagram form and deal with several abstract and concrete variables

Computer Skills
  • To perform this job successfully, an individual should have knowledge of the Microsoft Office Suite, DNA sequence analysis software (SnapGene, Vector NTI or similar)

Travel Requirements
  • The Scientist will be expected to travel to all applicable locations when needed

  • This position is located at the main headquarters located in Durham, NC and is required to physically be and work in the office, in order to be most effective, and at managers discretion
