The successful applicant will be responsible for performing studies on gene expression using immunohistochemistry (IHC) and RNA-scope (ISH), and then assembling target assessment packages that integrate protein/gene expression data from various other sources including RNA expression databases, Flow Cytometry analysis, and others. The goal of this work is to aid discovery and validation of candidate tumor targets for internal studies as well as work with collaborators. Therefore, the candidate must have a strong background in cancer biology, immunology and histotechnology. The candidate must also demonstrate a strong attention to detail, as the results of the studies conducted will play an important role in decisions to advance new therapies to the clinic for testing in patients. - Characterization of novel immunotherapeutic target expression by developing and performing both immunohistochemistry (IHC) as well as RNA scope (in situ hybridization) techniques, which involves the following below:
- Use literature and internal RNA expression databases to identify cell lines and tissues where targets are expressed
- Perform cell culture and FACS analysis to establish cell banks for use as positive and negative controls
- Validate IHC assays using both commercial and in-house antibodies on automated stainers, including:
Ventana Discovery XT, Leica BondRX, Dako Autostainer Plus
- Perform multiplex staining and potentially fluorescence staining as well
- Assemble image data in PowerPoint slides in a timely manner following staining
- Perform image analysis on stained tissues-can use HALO image analysis software as well as collaborate with Regeneron pathology team
- Clearly communicate results to management and external collaborators
