Summary
The position will primarily focus on the production of reagents associated with T-cell receptors. The individual will express recombinant HLA & TCR proteins in E. coli, & then refold & purify the resulting complexes. Additionally, the person will also express, purify, & characterize other recombinant proteins which support multiple targets within Regeneron's pipeline.

Responsibilities

• Express recombinant protein in E.coli, optimization of expression, & scale-up

• Cell lysis & clarification, isolation of inclusion bodies, solubilization of HLA-related proteins

• Solubilization & handling of peptides

• Refold & purify HLA/TCR complexes; biotinylation/conjugation of complexes, & labeling with fluorescent tags

• General protein purification with ÄKTA chromatography equipment & using affinity, ion exchange, size exclusion, & reverse phase methods

• Carry out experiments to improve protein solubility & stability

• Work collaboratively with other researchers to plan, update, & stay abreast of deadlines

• Documentation of experiments & results in an electronic LIMS notebook
Requirements

• 1-2 years applied experience with protein expression in E.coli & protein purification using affinity, ion exchange, reverse phase, & size exclusion techniques

• Experience running ÄKTA chromatography equipment

• SDS-Page electrophoresis, Western blotting, & spectrophotometry

• Sterile techniques

• Good Lab practices

• Data entry and notebook keeping using ELN

• Bachelors Degree required
