Summary
We are seeking a motivated research associate to join our Ophthalmology Department, devoted to the discovery and development of novel therapeutics for eye diseases. The ideal candidate has in vivo and in vitro expertise to help support target discovery, validation and new technology development. The candidate will provide scientific and technical support in the discovery, characterization and preclinical development of novel treatments for retinal degenerative diseases with focus in the RPE/choroid. A high level of technical expertise, such as confocal microscopy, immunohistochemistry, in situ hybridization and flow-cytometry are advantageous.
Responsibilities
The candidate will be responsible for the design and execution of experiments to generate in vivo and in vitro models of retinal degenerative diseases. The candidate will be involved in many aspects of drug development processes including target discovery, target validation, and assay development to support our Ophthalmology pipeline.
Lab duties for the candidate include, but are not limited to:
In vivo:
Animal (mouse, rat) handling, tissue dissection & collection
Perform SD-OCT in vivo eye imaging and functional behavioral tests and ERG
Apply or learn drug delivery and ocular procedures in rodents including intravitreal and subretinal injections.
Ex vivo molecular and cellular biology:
Protein and RNA extraction from various tissues, western blotting and RT-PCR.
Histological techniques (H&E, IHC, IF and ISH), RNAscope, image collection and quantitation.
Flow cytometry.
In vitro:
Cell culture and mainteinance of epithelial cells in transwell filters
Trans epithelial resistance measurements and surface-domain biotinylation of polarized cells.
Cell culture characterization using functional, molecular, and flow cytometry assays.
In addition to hands-on experimental work, the candidate will be expected to:
Perform advanced data analysis as well as troubleshooting with minimal supervision
Keep up-to-date and well-organized records of experiments
Communicate and report experimental findings at meetings.
Requirements:
Strong in vivo mouse experience including various drug delivery methods, characterization of disease models, tissue collection and dissection techniques
Experience with immunohistochemistry (protein or RNAscope) is highly desirable.
Basic molecular biology/biochemistry and cell biology methods including DNA and RNA isolation, PCR, genetic manipulation of cells (plasmid transfection, CRISPR editing).
Excellent interpersonal skills and ability to work in a team
Skilled in designing and executing well-controlled experiments that can provide reliable results
Minimum years of experience: 3-5 years of experience working with in vivo mouse models.
Demonstrated ability to collaborate with colleagues to support team-based projects.
Demonstrated ability to learn new knowledge and techniques
Troubleshooting, statistical concepts, and practical data analysis skills.
Preferred experience and skills:
Experience with ocular models of disease.
Basic knowledge of flow cytometry.
Good organization skills, excellent attention to detail, good interpersonal skills, perform as a team player.
Use of software for data analysis (Prism, Photoshop, Halo/ImageJ).
