Summary
The Genomic Engineering Technologies group is seeking an R&D Specialist to join in the effort to develop therapeutics for treating lysosomal diseases and other genetic disorders. This person will carry out cloning and molecular biology experiments to construct and validate potential biologic therapeutics including proteins and AAVs. This person will also assist in testing those molecules in vivo in mice and rats. The candidate should be highly motivated and be able to help design and carry out multiple mouse projects simultaneously.
Responsibilities
1. Carry out cloning and molecular biology experiments to construct and validate potential biologic therapeutics under the guidance of a Scientist. Desired skills include PCR, Gibson assembly, restriction digests, ligations, interpreting DNA sequencing results, and troubleshooting cloning experiments.
2. Evaluate potential biologic therapeutics in in vitro models. Desired skills include sterile cell culture, Western blotting, and ELISA.
3. Assist with in vivo experiments using mouse and rat models. Responsibilities will include injections by intravenous, subcutaneous, intraperitoneal, and intracerebroventricular routes, blood draws, and takedown and tissue harvesting. The ideal candidate will have some experience with rodents, but we will provide additional training as needed. 4. Manage mouse cohorts - monitor mice for health status, record clinical observations at regular intervals and carry out routine bio-fluid collections. 5. Carry out analysis of samples using techniques such as ELISA, SDS-PAGE/Western blotting, FACS/flow cytometry, and immunohistochemistry
6. Learn new techniques as needed
7. Keep a detailed lab notebook, updated in a timely and comprehensive manner
8. Present findings at group meetings, and format data for presentation to senior staff
Requirements
The candidate must have a bachelor's degree in a related field with at least 3 years of experience, or a master's degree with at least 1 year of experience.
Experience with molecular biology techniques such as Gibson assembly, PCR, cloning, ligation, transformation, transfection, and Western blotting is essential. Willingness to work with rodents in a laboratory setting is essential. Some prior experience working with rodents is essential.
The candidate should have a demonstrated ability to work independently with good organizational skills and attention to detail.
This position will require the individual to pass a physical before beginning employment.
