The department of PKDM supports the discovery and development of novel medicines by defining key pharmacokinetic, biotransformation and immunogenicity properties of drug candidates. Development of bioanalytical methods to quantify drug candidates and biotransformation products in biological fluids is therefore an essential part of PKDM's core mission.

The objective of this internship is to evaluate strategies for quantifying intact large molecule (LM) drugs by LC-MS/MS. Specifically the intern will evaluate different strategies for quantifying well characterized LM Amgen compounds or commercially available monoclonal antibodies (mAb) based drugs in biological fluids from humans or preclinical species. By nature of this investigation, the intern will have an opportunity to gain experience in immunoaffinity enrichment strategies on automated platforms, chromatographic separation of proteins and high resolution mass spectrometry, specifically using the Q-Exactive HF-X high resolution mass spectrometer (HRMS) platform.

Primary responsibilities would include:

  • Optimize HRMS and chromatographic parameters for intact LM bioanalysis
  • Optimize the immuno-affinity enrichment of intact mAb's from biological fluids from human subjects or preclinical species on the AssayMAP Bravo platform
  • Assess the use of internal standards for intact mAb quantification
  • Evaluate sample preparation strategies for intact protein quantification, specifically de-glycosylation and subunit generation (i.e., by IdeS/IdeZ proteases)
  • Comparing data quantification approaches (e.g., charge-state deconvolution, charge-state summing, and subunit quantification) for intact molecules
  • Optimize method conditions and assess sensitivity, linearity, precision and accuracy using calibration and QC samples generated in biological fluids
  • Results will be present internally (i.e., presentation and/or poster), and may be considered for external publication

Basic Qualifications

Amgen requires that all individuals applying for a grad internship or a co-op assignment at Amgen must meet the following criteria:

18 years or older

Graduated with a Bachelor's degree from an accredited college or university with a 3.0 minimum GPA or equivalent

Completion as a first year graduate student before internship starts and currently enrolled in a Master's program for a Masters internship OR a PhD for a PhD internship from an accredited college or university

Enrolled in an accredited college or university following the potential internship or co-op assignment

Must not be employed at the time the internship starts

Preferred Qualifications

Pursuing PhD with degree concentration(s) in Chemistry (Analytical, Biochemistry, or Bioanalytical)

Core focus in large molecule mass spectrometry (intact MS) or typical bottom-up LC-MS/MS approaches

Direct experience using HRMS platforms, especially Orbitrap based systems
