The Senior Research Investigator will join the Mechanistic Disease Discovery Research team at Bristol-Myers Squibb in Redwood City CA to provide leadership for a group focused on rapid next-generation sequencing (including single cell) on critical human samples (immune, tumor, and stromal cells) as well as head CRISPR and gene expression efforts for the site.

The qualified candidate is a highly motivated, interactive, and creative individual that possesses substantial experience in end-to-end next-generation sequencing and will be responsible for building capability for the site in Redwood City. The candidate will collaborate closely with other Discovery sites within BMS, providing a high-visibility opportunity for career development. The candidate will also be responsible for heading the CRISPR and gene expression efforts at the site utilizing cellular and molecular biology to engineer and characterize cells expressing or suppressing gene expression through recombinant methods. The candidate will be expected to effectively collaborate with colleagues in multiple groups and will have the opportunity to generate significant and impactful data on human tissue analysis. In this capacity, the candidate will be expected to contribute scientifically to these efforts. The successful candidate has superb verbal and written communication skills and is capable of presenting scientific results to multidisciplinary teams, senior management and at external scientific meetings. The candidate will act as an expert resource in his/her scientific field and in related disciplines, and collaborate across a highly matrixed organizational structure.

Responsibilities:

• Build a NGS lab end-to-end and operate this core function to support site sequencing needs

• Generate data from NGS (including single cell) and managing data flow

• Manage a team executing on the NGS and gene manipulation activities for the site

• General molecular biology involving isolation, manipulation, and characterization of DNA, RNA and proteins

• Design of mammalian cell expression vectors for recombinant proteins, inhibitory RNAs, and CRISPRs

• Mammalian cell culture including hematopoietic cells and cell lines, including those of rodent or human origin

• Mammalian cell manipulation including transfection or lentivirus transduction of expression, interfering RNA, or CRISPR vectors

• Assessment of RNA and protein expression using qRT-PCR, flow cytometry, ELISA, HTRF or western blotting as appropriate

• Communication of work, plans and progress using written and verbal communication in small and group settings

Qualifications:

• PhD degree with 4+ years of relevant postdoctoral experience in NGS and cellular/molecular field of practice. Experience in the field of immunology or oncology is ideal.

• Extensive experience in NGS; experience in managing a NGS facility is a strong plus. Experience in managing NGS data flow.

• Management/supervisory experience is required.

• Demonstrated experience in design and construction of cell expression plasmid constructs, site-directed mutagenesis, and recombinant protein vector design.

• Demonstrated experience in pooled or arrayed CRISPR library generation and screening, including downstream library preparation next gen sequencing is ideal.

• Extensive cell culture techniques including demonstrated ability to establish transient/stable cells and cell lines through transfections and/or virus transduction. Experience in work with primary human or rodent cells.

• Expertise in characterization of protein expression through biochemical (ELISA/Western) and cellular analysis (binding,reporter analysis) and RNA expression using RT-PCR. Flow cytometry experience is a plus.

• Ability to multitask among multiple projects and teams and work independently in a fast-paced, highly interactive environment.

• Excellent oral and written communication skills.
