Project Description: The First goal of this intern project is to create a universal small-scale column packing method to build dynamic small-scale packing procedures. The second goal is to examine the current capabilities of process analytical technologies to differentiate between poor column packs and process variability on those packed columns.

With the lack of understanding of packing parameters, resources and efficiency is wasted into packing a small-scale column. The intern will responsible for packing small-scale chromatography columns using various packing methods. Different packing parameters (i.e. slurry concentration, compression factors, packing flow rates, buffer types, different resins, and column sizes) will be analyzed to create a universal small-scale column packing method with a high level of repeatability.
The second part of the project will include performing chromatography runs on each column to model manufacturing column stresses. A control column will be packed to manufacturing specifications; this column will be processed using varying parameters such as column loading to produce atypical chromatographic results. Transition Analysis will then be used to differentiate between columns experiencing atypical features due to packing inconsistencies, and atypical features due to the varying processing parameters.
Desired Majors and/or Skills:

• Chemical Engineering major.

• Experience with chromatography/protein purification.

Desired Education Level:
- Rising Junior, Rising Senior, or Master's in Engineering Candidate.
