Position Summary

Known for its scientific and operational excellence, Regeneron is a leading science-based biopharmaceutical company that discovers, invents, develops, manufactures, and commercializes medicines for the treatment of serious medical conditions. Regeneron commercializes medicines for eye diseases, high LDL-cholesterol, atopic dermatitis and a rare inflammatory condition and has product candidates in development in other areas of high unmet medical need, including rheumatoid arthritis, asthma, pain, cancer and infectious diseases.

These positions are in Protein Chemistry in Tarrytown, NY. There are 4 openings for Summer 2018. Each opening is described below.

Position 1: The student will gain hands-on-experience in bioconjugation of critical protein reagents with a focus on optimizing the steps in bioconjugation. The student will also perform characterization of bioconjugated proteins using various biophysical and biochemical characterization techniques. The bulk of the internship will focus on evaluating the effect of pH on stability, conjugation ratios, formulation conditions, buffer exchange vs dialysis and other factors that impact the quality of the conjugated protein. The findings from these studies will be summarized and presented to a larger group at the end of the project.

Position 2: The student will have the opportunity to acquire skills and hand-on experience in the area of protein characterization with focus on Western Blotting (WB) and Capillary Electrophoresis (CE). The student will be trained to optimize CE based WB method to detect the specific bands in bispecific antibodies and monoclonal antibodies. The main aim of the project will be to optimize the detection conditions for different REGN antibodies. The ideal candidate will be efficient, hard working, detail oriented and able to perform successfully in a team environment. Upon successful completion of the project, the student's work will be eventually applied to help characterize proteins to support the preclinical development of novel antibodies.

Position 3: The student will gain hands-on experience in protein characterization using various analytical techniques and provide exposure to experimental design. Project will investigate the sensitivity of select analytical assays currently used in Regeneron reference standard extended characterization. The goal is to do systematic forced degradation or spiking studies at various levels and observe when we begin to see differences from control sample by each assay. The project may also involve systematic study of effects of excipients on assay sensitivity and exploring alternative ways to remove excipients such as polysorbates without affecting product profile. Intern will first train on the analytical techniques and then perform testing on selected REGN antibodies. The ideal project outcome will give insight on the analytical power and limitation of our assays in monitoring REGN reference standard stability.

Position 4: The student will focus on investigating the effect of common environmental stresses (eg. oxidation, deamidation, light, etc.) have on high order structure in monoclonal antibodies through the assessment of antibody structural content change. The project will also encompass a secondary goal of applying QbD and DoE concepts in evaluating the sensitivity, reproducibility, and methodology robustness of various high order structure techniques & instruments (eg. Capillary DSC, FTIR).

Initial efforts will encompass generation of stressed materials through various mechanisms (eg. oxidation, deamidation, etc.) and fractionation of these materials by size or charge. These different environmental stresses are expected to impact mAb domain structure and function (i.e. Fab & Fc) differentially, and HOS techniques such as DSC will be employed to assess the nature and degree of these impacts. These assessment will inform future interpretation of data from DSC and other HOS analytics, as well as how to relate these interpretations to critical quality attribute and give direction for development of future methodologies.

The secondary project goal will be the evaluation and development of our various HOS techniques and instruments using a QbD framework, thereby assessing accuracy, linearity, robustness, repeatability, range, and LOD/Q. The evaluation will tentatively be focused on certain high order structure techniques and the corresponding instruments, including FTIR for our secondary protein structural technique and DSC for our secondary/tertiary protein structural determination.

• Must be an undergraduate or graduate student
• Must be pursuing a degree in Biochemistry, Biotechnology, Biological Sciences or related
• Position 1: Experience in a laboratory setting for 6 months to a year, as well as experience with pipetting and buffer preparation and Some knowledge related to biotherapeutics
• Position 2: Experience with SDS-PAGE, western blot, and Capillary electrophoresis
• Position 3: Experience with SEC, Flurescence, FTIR, and Circular Dichroism (CD)
• Position 4: Experience with SDS-PAGE, protein purification, SEC & ion-exchange Chromatography, Differential Scanning Calorimetry (DSC) is preferred.
• Must have strong attention to detail. Must be well organized. Must have strong presentation skills. Must have the ability to balance multiple experiments. Must have strong time management skills.

• Must be enrolled in, or accepted to, an academic program pursuing a Bachelor's, Master's, PhD or PharmD
• Summer Program is full-time (~40 hrs/wk) for at least 10 weeks and is paid
• Prefer demonstrated leadership in areas such as campus activities, clubs, sports or the community
• Minimum GPA of 3.0
• You will work with a specific manager on a project(s)
