Position Summary

Known for its scientific and operational excellence, Regeneron is a leading science-based biopharmaceutical company that discovers, invents, develops, manufactures, and commercializes medicines for the treatment of serious medical conditions. Regeneron commercializes medicines for eye diseases, high LDL-cholesterol, atopic dermatitis and a rare inflammatory condition and has product candidates in development in other areas of high unmet medical need, including rheumatoid arthritis, asthma, pain, cancer and infectious diseases.

This project will support a bispecific ADC program within the Oncology-Angiogenesis department in Tarrytown, NY.
• Perform in vitro binding, internalization, receptor degradation and cell killing assays in cancer cell lines. This will help in the selection of tumor models in which to study the bispecific-ADC anti-tumor efficacy.
• Determine expression of the bispecific ADC targets in PDX tumors from commercial sources. This will help to identify PDX models in which to study the anti-tumor efficacy of the bispecific-ADC.
• Determine expression of one of the bispecific-ADC targets in organs harvested from humanized mice.
• Assist in the execution of an arrayed CRISPR screen aiming to elucidate the machinery responsible for the internalization of the bispecific-ADC. Particularly, perform internalization assays.
• Perform internalization assays in cells transfected with mutants of the bispecific-ADC targets. These constructs (14 already generated) bear mutations in candidate endocytic signals of the targets.
• Assist in in vivo studies (i.e. process serum samples for ADC concentration determination).
