Description


This position provides scientific and technical support to Core Sciences & Technologies and other relevant departments at Takeda California in the areas of molecular biology and protein expression. The candidate is expected to contribute in the following areas:

*   Design and expression of various secreted or membrane-associated proteins to support in vitro screening and in vivo research studies
*   Optimizing expression of TCAL candidate mAbs to support antibody discovery through clinical entry

Requirements


*    Provide protein expression leadership within the Cell Engineering and Molecular Biology group. This will involve managing all molecular biology and expression  activities within the group, including:
*    Collaborating with colleagues in Research to design and engineer protein sequences (full-length, fragments, mutants, fusion proteins) appropriate for their intended use
*    Expressing a variety of proteins (mAbs, Fc- fusion proteins, antigens, and protein domains) in secreted or membrane-bound form using mammalian (i.e. CHO or HEK293)and non-mammalian (i.e. yeast or ecoli) expression systems.
*    Leading efforts to optimize transient expression of antibodies to support antibody discovery and preclinical studies
*    Leading efforts to optimize expression of antibodies (transient and stable) to support antibody discovery and preclinical studies. Specific activities could include:

         * Designing expression constructs for lead antibodies, also identifying potential manufacturing liabilities
         * Evaluating vector elements (i.e. promoters, signal peptides, etc.) or cell selection strategies to enhance           transient and stable antibody expression
         * Providing technical leadership in cross-functional teams across the global Takeda organization to                     identify and implement best practices for antibody expression      

*   Actively survey industry-standard and cutting-edge platforms for protein expression either through participation in workshops or conferences
*   Managing and providing technical guidance to junior staff members within the group

EDUCATION, EXPERIENCE, KNOWLEDGE AND SKILLS:

*   PhD degree in life sciences or equivalent with Post Doc and 0-4+ years of relevant experience, preferably industry; MS/MA in life sciences or equivalent with a minimum of 10 years of relevant experience; BS/BA in life sciences or equivalent with a minimum of 12 years of relevant experience
*   Experience and knowledge in recombinant DNA techniques and protein engineering to design, construct and optimize expression vectors for a variety of proteins in both mammalian (CHO and HEK293) and non-mammalian systems (ecoli or yeast) is required.
*   Familiarity with flow cytometry and/or sorting is preferred
*   Knowledge of the Regulatory standards for expressing therapeutic mAbs a plus
*   Excellent written and verbal communication skills, along with the ability to work in cross-functional project teams
*   Demonstrated success as a leader, managing people and projects
