Position Summary

The candidate will play a lead role in Protein Sciences, with responsibilities to include the supervision of protein purification and analytical characterization for both small and large scale antibody and antigen production.  Both within the Protein Sciences research group and within interdisciplinary project groups, the candidate will provide technical and scientific oversight to solve challenging problems, speed project timelines and potentially reduce needed resources. The incumbent  will  coordinate protein expression, purification, and analytics activities and will supervise the day-to-day and long-term goals and activities of three or more staff.

Essential Functions

•   Play a leadership role in the expression, purification and analytical characterization of recombinant proteins
•   Responsibilities of Protein Sciences group:
     o   Design and generate therapeutic antibodies, antigens, and secondary reagents
     o   Develop and implement methods for purification and deep biochemical and biophysical characterization of therapeutic candidates and key reagents, and develop methods for identifying candidate antibodies with potential to be formulated at high concentration.
     o   Transient and stable protein expression in mammalian cells
     o   FPLC purification of proteins and reagents
     o   HPLC analysis of purified recombinant proteins and peptides
     o   Protein quantification and characterization assays (SDS PAGE, IEF, etc.)
     o   Non-specific labeling of proteins
     o   Analytical analysis of protein thermal stability using thermofluor/lightcycler and DSC technologies
•   Provide technical oversight and leadership in the purification and in-house development activities for antibody lead candidates including formulation and preclinical activities
•   Participate in multi-departmental research and development projects and discussions, providing scientific leadership and direction regarding activities related to Protein Sciences
•   Act as in-house expert in matters related to protein chemistry and purification instrumentation, technical, or scientific areas (e.g., HPLC, FPLC, DSC, etc.).
•   Pro-actively assist others within Protein Sciences in solving technical problems.
•   Partner with Development and Manufacturing to ensure seamless transition and continued support, when necessary, of therapeutic candidates.
•   Coordinate experiments with colleagues to ensure rapid achievement of objectives and overall goals.
•   Manage the day-to-day and long-term activities, objectives and performance of staff.  
•   Train staff in core functions as needed.
•   Proactively use literature to investigate and recommend new instruments and/or methods, assays, or project targets.
•   Implement new methods, screening, and protocols as needed.
•   Direct process improvements for the group.
•   Ensure data generated is well-controlled and interpreted appropriately. Communicate findings, conclusions and potential strageties openly and effectively to colleagues and senior management.
•   Maintain an up-to-date electronic laboratory notebook


•   Lead AnaptysBio projects with the goal of rapid identification of superior therapeutic candidates, to include:
     o   Conceive strategies for developing new drug concepts through advancement of candidates to clinical development.
     o   Lead project team meetings and coordinate activities for the project team with other research groups.
     o   Write project reports and lead program reviews
     o   Perform and communicate background research within the therapeutic/project area, including design goals, lead identification strategy, competitive analysis and IP.
•   Stay current with the relevant scientific literature, and present and explain key concepts to colleagues.
•   Write and review manuscripts and intellectual property documents.
•   Participate in industry conferences and seminars with occasional travel required.
•   Follow and adhere to SOP’s and laboratory safety regulations.

Requirements


Job Requirements and Qualifications

Education & Experience

•   PhD biochemistry, bioengineering, biophysics or related field with post-doctoral experience specializing in protein science, molecular biology, or related.
•   10+ years, post-doctoral, of relevant experience with a minimum of 6 years industry experience in biotech and/or pharma.
•   5-8 years people management experience within a laboratory setting
•   Extensive background in protein chemistry, including protein purification, protein analytics, biophysical properties optimization, protein structure and function, labeling and/or alternative chemical modification techniques, antibody generation and characterization, including humanization and affinity maturation.  
•   Experience with screening technologies and their integration into the drug discovery and development process.
•   Proven experience leading  research programs aimed at identifying and optimizing candidate therapeutic proteins (preferably antibodies).
•   Demonstrated experience managing a group of protein scientists involved in protein engineering, characterization of proteins and labeling technologies.

Knowledge Requirements

•   Expertise in the production and analysis of recombinant proteins to therapeutic standards, including application of state of the art bioanalytical methods for quality assessment.
•   Expertise in all aspects of antibody engineering, including humanization, sequence and affinity optimization, and design of multispecific antibodies.
•   Proficiency working with the Microsoft Office suite programs (Project, Excel, PowerPoint, Word, etc.).
