Construction of expression vectors. Generation of proteins in mammalian cells. Single cell cloning and selection of clones with high titer.
Generation of rodent monoclonal antibodies including screening hybridoma arrays, and in antibody/protein engineering including expression in mammalian cells.  Experience with a broad range of assays involving different human cell types including gene expression, immunohistochemistry, FACS, immune cell assays and other cell-based assays will be an advantage.
Responsibilities:
Construction of expression vectors. Generation of stable cells. purification of expressed proteins. Functional studies of expressed proteins in biochemical and cell based assays.
Assays for protein analysis including but not limited to ELISA, HPLC, protein-protein interaction.
Preparation of protein antigens for immunizations including generating fusion proteins and cell lines expressing surface antigen.  Generation of hybridomas from immunized rodents and screening of hybridoma arrays for antigen binding, cell binding and other properties.
Cloning of antibody genes and expression in mammalian and other cell types.  Modification of antibody genes, for example for humanization and generation of single chain antibodies.  Testing of antibodies in different assay types including binding, in vitro and in vivo assays, and immune cell assays especially with T cells.
Input into publications, patents and regulatory submissions, and participation in internal and external scientific meetings.
