Job Description

Position Summary

This position is responsible for cell line generation and maintenance for recombinant protein production (expression, scale-up and purification of proteins), and in doing so will lead protein expression efforts of a small group of scientific staff.

Essential Functions

•   High-throughput transient monoclonal antibody (mAb) expression from 96-well plates to shake-flask cultures
•   Stable cell line and stable pool generation, scale up & maintenance (including but not limited to HEK293 and CHO) for recombinant protein and antibody production
•   Large-scale fed-batch culture of stable CHO clones and pools to provide gram-scale quantities of mAbs for further analysis
•   Transient transfection of HEK293 and CHO cells for routine recombinant protein production
•   Optimize expression in both transiently-transfected and fed-batch cultures
•   Optimize media and methodology used in protein production
•   Run routine immunoassay procedures for mAb quantitation from cell culture supernatants, and develop new assays as needed
•   Analyze experimental data and present findings/recommendations at internal project and department meetings, contributing to the advancement of project objectives
•   Contribute to external technology transfer efforts related to cell culture and expression activities
•   Supervise and mentor junior scientific associate staff
•   Maintain organized and accurate records/lab notebooks, including the creation of new SOP’s where appropriate
•   Maintain familiarity with current scientific literature, actively searching literature to identify alternative methodologies and solve scientific problems; implement new techniques and technologies found in literature as appropriate.
•   Coordinate overall workload and workflow of Protein Expression group including creation of work plans for evaluation of new program targets and developing manufacturability assessments for lead antibodies
•   Independently design, execute and interpret experimental studies for cell line generation and maintenance for recombinant protein production.
•   Independently establish and validate new methodologies and experimental designs.
•   Contribute scientific knowledge to the development of concepts, and to the achievement of objectives.
•   Take initiative in projects by solving problems, providing direction and/or assistance to junior staff, and implementing new procedures.

Additional Responsibilities:

•   May be responsible for identifying patentable inventions
•   Follow and adhere to SOP’s and laboratory safety regulations
•   May write research study reports, sections of manuscripts, patent applications, and/or contribute to scientific journals.
•   Contribute to the general laboratory environment, act a good steward/citizen and suggest and promote good laboratory policies.



Education & Experience:

•   BA/BS or MA/MS in Biology or related field
•   Bachelors’ degree in Biological Sciences (or related degree) and least 8+ years related experience in role(s) of increasing independence and responsibility; or Masters’ degree and 5+ years experience.   Preference will be given to candidates with significant experience in cell line development, mammalian cell culture process development and bioproduction. Experience within the field of antibody research is a plus.
•   Experience with WAVE bioreactors
•   Experience and knowledge of Design-of-Experiment methodology, including fractional factorial analysis and mixing studies
•   Experience with CHO stable cell line generation & screening for optimal expression by ELISA is a plus
•   Experience with other cell types (bacterial, yeast, baculovirus) for protein production is a plus but not required
•   Experience with vector construction is a plus
•   Experience with protein purification (FPLC) and analysis (SDS-page, Western blot, HPLC) is a plus
•   Proven track record of scientific accountability and clear ownership of responsibilities

Knowledge Requirements:

•   Strong knowledge of protein purification, biochemistry and at least some exposure to assay development
•   Strong scientific knowledge of recombinant protein expression
•   Mammalian cell culture and cell transfection methodologies
•   Proficiency working with Microsoft Word, Excel, and PowerPoint
•   Proficiency with sequence analysis software is preferred, including sequence alignments, structure prediction and similarity searches
