Job Description


Position Summary:

We seek a highly motivated and experienced protein biochemist to join the Therapeutic Antibody group at Achaogen to make significant contributions to drug discovery projects targeting disease areas with high unmet medical need. The successful candidate will operate in a highly diverse and dynamic research environment supporting early research and drug discovery programs. They will use their expertise to advise and support therapeutic projects through the expression, purification and characterization of high quality recombinant proteins and antibodies. Attention to detail, the ability to communicate across multiple scientific disciplines and a persistent drive for results are critical to the role.

Essential Functions:
•   As an integral member of drug discovery project teams, provide expertise and guidance on protein design and production strategies.
•   Utilize antibody engineering approaches to fit therapeutic format to desired functional properties on a project-to-project basis.
•   Express, purify and characterize antibody candidates derived from internal discovery platform at small- and mid-level scale; contribute to the creation of novel antibody formats for therapeutic application.
•   Perform biophysical characterization studies of antibody candidates, including epitope binning & affinity determination using Octet or Biacore systems.
•   Employ cell culture expertise to generate cell lines that stably or transiently express soluble or cell surface proteins for use in antibody discovery and functional characterization activities.
•   Independently operate analytical instruments such as spectrophotometers, analytical FPLC (AKTA Pure), plate readers, light scattering systems.
•   Develop and implement creative strategies for high throughput solutions and optimize workflow of existing methods for Achaogen’s protein expression, purification and characterization needs.

Requirements


Essential Experience:

•   Expertise in the use of mammalian and/or insect cell expression platforms, purification systems and analytical instruments (e.g., HEK293 and/or CHO transfections, AKTA/FPLC).
•   A proven record of success in the characterization of mammalian proteins, including antibodies, antibody fragments, and novel protein constructs.
•   Hands-on experience with antibody engineering approaches to harness optimal therapeutic functionalities or to solve potential manufacturing liabilities.
•   Demonstrated ability to enhance protein production capabilities through identification of key technology improvements, innovation or problem solving.
•   Experience with automation, high-throughput techniques and robotics.

Preferred Experience:

•   6+ years of research experience in an industry or academic setting with an emphasis on protein and antibody production/purification/analysis.
•   Experience in the expression, purification and characterization of bispecific antibody constructs and other non-standard protein formats is a plus.
•   Demonstrated success in a dynamic research environment where adaptability to new projects and approaches was essential.
•   Antibody binding characterization using Octet or Biacore instrumentation.

Education:

•   Minimum of a BS or MS degree in a relevant scientific field (e.g. biochemistry or molecular biology).
