Description

Sutro Biopharma, located in South San Francisco, is developing a new generation of antibody drug conjugate therapeutics and bifunctional antibody-based therapeutics for targeted cancer therapies. These therapeutics will significantly extend the clinical impact of current oncology therapeutic approaches and are beyond what can be envisioned with current cell-based expression technologies. Sutro's biochemical synthesis technology, which underpins these therapeutics, allows the rapid and systematic exploration of many protein drug variants to identify drug candidates. Once these product candidates are identified, production can be rapidly and predictably scaled up to commercial levels. Sutro has established a Good Manufacturing Practice (cGMP) facility for the production of clinical supplies of materials using its biochemical protein synthesis platform. Sutro Biopharma is collaborating with pharmaceutical and biotech companies in the discovery and development of novel protein therapeutics.

Sutro Biopharma, Inc. is looking for an energetic and talented individual to join the multidisciplinary Protein Biochemistry group within the Research Department. This position will be working to understand and improve the performance of our core in vitro biochemical protein synthesis technology by designing and executing experiments on transcription, translation, protein folding, purification and analysis.  

Responsibilities:
You will participate in cross-functional teams and collaborate on multiple projects. Responsibilities extend from gene design through protein expression, purification and characterization. Protein expression is conducted in a number of different formats, from simple unmonitored static and shaking systems to more complex single or parallel systems using computer monitoring and feedback control and in scales ranging from microliters to several liters. The successful candidate will design & validate protein expression and purification methods appropriate to the scale, number-of-samples, and required purity level for the given project, and will implement appropriate assays to characterize product and to better understand the technology and process. In addition, the successful candidate will be required to interface with external collaborators, set-up and run team meetings, make presentations to outside collaborators and senior management, and to author internal and external publications.

Requirements

We are seeking a highly-motivated team-player with excellent problem-solving skills and experience in protein production, purification and analysis. Versatility is a must, as tasks will involve multiple different protein types, scales and purity requirements. Good organizational skills and the ability to manage projects within a timeline are essential.

The ideal candidate will have the following background/skill set:
• Ph.D. in Biochemistry or a related discipline, with 5+ years industry experience.
• Experience working with antibody expression is required.
• Hands on experience with WAVE and stirred tank bioreactors is desired.
• Experience with antibody analytical methods is required. In particular, HPLC analysis using SEC, HIC, CIEX or RP. Analysis of binding affinity using Forte-Bio or Biacore, and set-up of activity assays is a plus.
• Proficiency with common protein analytical techniques including SDS-PAGE, Western blotting and ELISA.
• Proficiency with techniques used in gene synthesis, including primer design, PCR and subcloning.
• Familiarity with sequence analysis software, such as Vector NTI.
• Hands on experience with various research-scale affinity methods for protein purification, including nickel IMAC, protein A, protein G etc. Experience in parallel processing of samples for purification using Kingfisher preferred. Experience with Akta systems a plus.
• Experience working with radioisotopes is preferred.
• Experience working on cross-functional teams and managing others are pluses.
