The Biotechnology Discovery Research (BioTDR) organization in Eli Lilly & Company is responsible for discovering and optimizing novel biotherapeutics (peptides, proteins, antibodies) for advancement into the clinic in key areas of unmet medical need including diabetes, immunology, oncology, neuroscience, and others. The BioTDR organization has recently created the Formulation & Delivery Team (FDT) focused on establishing capabilities in the areas of physicochemical characterization of biologics, formulation development, and early assessment of device compatibility. This team works cross-functionally with both discovery and development groups including protein engineering, therapeutic area biology, ADME, chemistry, and bioproduct development teams to ensure that compounds entering development have characteristics that will ensure successful translation from discovery to the clinic. The Oral Peptides Delivery Group is looking for a talented, highly-motivated, and technically competent scientist to become part of our team.
The key purpose of this position is to support discovery and development of novel peptides with ideal molecular attributes to allow their oral delivery. By establishing a battery of high throughput assays that assess the physical and chemical properties of peptides, the ideal scientist will provide critical data that define the molecular attributes of drug candidates and allow the development of stage-appropriate formulations for preclinical studies. The ideal scientist is expected to work effectively with partners from peptide chemistry, biology, and ADME to support drug discovery from hit identification to early phases of clinical development. The selected scientist will also be responsible for leading/supporting projects and organization initiatives and bringing new capabilities/technologies to the Oral Peptides Delivery Group.
The responsibilities for this role will include:
• Identify/establish screening assays to characterize the physical/chemical properties of peptides/proteins that influence oral absorption
• Perform analytical method development for peptides/proteins using multiple analytical/spectroscopic tools
• Design and perform studies to characterize the physicochemical properties of peptides/proteins, analyze the results, and examine the correlation(s) with molecular structure, in vitro permeability, and in vivo oral bioavailability
• With support from the automation group, transfer benchtop assays to robotic screening platforms; integrate the assay platform, supporting analytical tool, and data analysis systems to achieve high throughput capacity
• Troubleshoot protocols, critically analyze data, and present screening results at project team meetings
• Keep abreast of relevant new technologies/capabilities relevant to this role
• Author data summaries, technical reports, standard operating procedures, publications, and presentations
Basic Qualifications •PhD in Biophysics, Chemistry (Analytical, Bio, Organic, Physical), Chemical Engineering, Pharmaceutics, Pharmaceutical Sciences, or other related fields with relevant expertise
•Relevant postdoctoral experience in academia, national/government labs, or pharmaceutical/biotech industry for 1-5 years
Additional Skills/Preferences •A solid research record demonstrated by patents, publications, or presentations in the area of peptide characterization, assay development, bioanalysis
•Interest and ability to work in a dynamic, fast-paced environment, and be capable of handling multiple tasks while meeting timelines
•A strong understanding of peptide/protein chemistry, structure, conformation, physicochemical properties and the relationship to solution behavior, self-association, aggregation, chemical and thermal stability
•Experience with a range of biophysical and chemical analytical techniques (HPLC, AUC, dynamic and static light scattering, CD, ITC, SPR assays, spectroscopic methods) that can be applied for thorough characterization of peptides and proteins
•Foresight to integrate multiple orthogonal in silico and in vitro assays to address project’s critical needs
•Demonstrated expertise in high-throughput assay development, implementation, and data analysis
•Prior experience with automation and liquid handling platforms is highly desirable
•Ability to independently contribute with minimal supervision from the bench (design and execution of studies) to inform decisions in a cross-functional team
•Ability to lead projects and work productively in an interdisciplinary team environment
•Excellent oral and written communication skills; Experience with data management software
•Flexibility to adjust priorities over time to meet organizational and scientific goals
•Some experience with supervision of direct reports
