“This article demonstrates the importance of understanding the mechanism of action of phosphodiesterase inhibitors and so our new methods for enriching phosphodiesterases from natural sources will improve the quality of research in this area. However, because of the instability of cyclic nucleotides, classical substrate affinity enrichment is not an option for these enzymes, so we worked around that problem to develop PDEnrich™. As a result, this product can be a crucial first step to prospect into the structure and function of phosphodiesterase variants.” states Swapan Roy, Ph.D., President of Biotech Support Group.
PDEnRich™ is derived from a silica-based NuGel™ multi-dimensional surface library and optimized for the enrichment of phosphodiesterase activity. The standard prep protocol starts with 100 µl of tissue/cell extracts, or approximately 0.5 mg total protein, but the process can be scaled up or down to accommodate different sample volumes and protein concentrations. The kit includes all necessary reagents for immediate use.
